AB  - Methods of absorption and Raman spectroscopy were applied in this study to analyze functions and amounts of photosynthetic pigments in the leaf of maize inbred lines (M1-3-3Sdms, ZPPL186, and ZPPL225). Absorption spectroscopy was used to determine the dynamics of biosynthesis of chlorophyll a, chlorophyll b, and carotenoids. Raman spectroscopy was used to present the dynamics of biosynthesis of carotenoids and other compounds in terms of origin and kinetic form of the formation of spectra with all spectral bands. Organic molecules and compounds causing the formation of certain spectral bands in the Raman spectrum (carotenoids, phosphates, glycogen, amid III, and others) were determined. Conformational and functional changes of photosynthetic pigments in the leaves of the maize inbred lines, which occur due to changes in the ratio (quotient), were analyzed. The ratios (quotients) obtained indicate different contributions of valence oscillations of their chemical bonds, which inevitably alter the conformation of molecules and compounds. The results presented for the overall study point to minor biogenic differences among the maize inbred lines under study.
AB  - У овом раду примењене су методе за апсорпциону и Раманову спектроскопију у изучавању функција и количина фотосинтетских пигмената у листу инбред линија кукуруза: M1-3-3Sdms, ZPPL186 и ZPPL225. Помоћу апсорпционе спектроскопије утврђена је динамика биосинтезе свих форми хлорофила: хлорофил a, хлорофил b, хлорофили a + b и каротеноида. Помоћу Раманове спектроскопије показана је динамика биосинтезе каротеноида и других једињења у погледу места и кинетичке форме настајања спектара са свим спектралним тракама. Утврђени су органски молекули и једињења који условљавају настајање појединих спектралних трака у Рамановом спектру (каротеноиди, фосфати, гликоген, амид III и други). Анализиране су структурне и функционалне промене фотосинтетских пигмената у листу инбред линија кукуруза до којих долази због промене односа (количника). Добијени односи (количници) указују на различите уделе валентних осцилација и њихових хемијских веза, што неминовно мења конформацију молекула и једињења. Изложени резултати указују на мање биогене разлике између проучаваних инбред линија кукуруза.

AB  - В настоящее время мало неинвазивных методов, которые позволяют контролировать со-держание и конформацию молекул биоактивных органических соединений в клетках и тканях рас-тений не только в лабораторных, но и в полевых условиях. Инфракрасная спектроскопия и спек-троскопия комбинационного рассеяния (ИК- и КР-спектроскопия) активно используется для ана-лиза роли молекул некоторых веществ в селекции сельскохозяйственных культур. С помощью ме-тодов вибронной спектроскопии мы исследовали изменения содержания и конформации молекул хлоропластов у различных линий кукурузы. Методом ИК-спектроскопии (область 3500-3000 см−1) установлено, что в воде, углеводах и белках хлоропластов доля колебаний ОН-групп и внутри- и межмолекулярных Н-связей максимальна у линии кукурузы ZPPL 186, а колебаний NH-групп ами-дов (белки) минимальна у линии ZPPL 225. Доказано, что для линии ZPPL 186 характерна максимальная доля колебаний от молекул алканов, карбоновых кислот (область 2920-2860 см−1) и деформационных колебаний ароматических структур (полоса 1000 см−1), находящихся в хлоропла-стах, тогда как для линии M1-3-3-sdms — доля валентных колебания O=C=O связей (полоса 2300 см−1). С использованием КР-спектроскопии (области 1250-500 см−1 и 1535-1400 см−1) обна-ружено, что различия в спектрах между линиями кукурузы связаны с выявленными изменениями конформации молекул каротиноидов в хлоропластах, но не молекул целлюлозы. Установлено, что у всех образцов (кроме ZPPL 225) молекулы каротиноидов находятся в 15-trans форме с различной конформацией полиеновой цепи. Отметим, что у линии ZPPL 186 конформация каротиноидов, находящихся в хлоропластах, характеризуется минимальной величиной поворота вне плоскости полиеновой цепи и более выраженными колебаниями боковой СН3-группы. Высказывает пред-положение, что у изученных линий кукурузы каротиноиды хлоропластов листа не взаимодействуют с ароматическими аминокислотами белков. По мнению авторов, сочетание методов ИК- и КР-спектроскопии хлоропластов листа можно рекомендовать для контроля за содержанием и конфор-мациоными изменениями биомолекул при селекции кукурузы.
AB  - Currently, there are few non-invasive methods that allow you to control the content and conformation of molecules in plant cells and tissues, not only in the laboratory, but also in the field. Infrared microscopy and Raman spectroscopy (IR and Raman spectroscopy) are actively used to ana-lyze the role of molecules of certain substances in crop breeding. Using methods of vibronic spectroscopy, we investigated changes in the content and conformation of chloroplast molecules of various maize lines. Using infrared (IR) spectroscopy (3500-3000 cm−1) it was found that in the chloroplasts, the proportion of vibrations of OH-groups and intramolecular and intermolecular H bonds is maximum for the maize line ZPPL 186, and vibrations of NH-groups of amides (proteins) is minimal for ZPPL 225. It has been proven that ZPPL 186 chloroplasts are characterized by the maximum proportion of stretching vibrations from alkane molecules, carboxylic acids (region 2920-2860 cm−1) and deformation vibrations of aromatic structures (band at 1000 cm−1), and for the line M1-3-3-sdms, the fraction of stretching vibrations of O=C=O bonds (band at 2300 cm−1) is characteristic. Using Raman spectroscopy (range of 1250-500 cm−1 and 1535-1400 cm−1), it was found that differences in the chloroplasts of different maize lines are associated with changes in the conformation of chloroplast carotenoid molecules (rather than cellulose molecules). It was found that in two samples (except ZPPL 225), carotenoid molecules are in the 15-trans form with different conformation of the polyene chain. We note that the conformation of carotenoids of the ZPPL 186 line is characterized by a minimum amount of rotation outside the plane of the polyene chain and has more pronounced vibrations of the lateral CH3-group. It was assumed that carotenoids of leaf chloroplasts of various maize lines lack interactions with aromatic amino acids of proteins. According to the authors, the combined use of IR and Raman spectroscopy of the leaf chloroplast fraction can be recommended for monitoring the content and conformation of biomolecules in maize breeding.

Identification of characteristic organic molecules in kernels of maize (Zea mays L.) hybrid grain using infrared spectroscopy

AB  - Modern biophysical methods have provided a breakthrough in investigations of the status and functions of the intact plants at the molecular level. The infrared (IR) spectroscopy allows us to analyze the molecular composition and structure by recording the absorption of infrared radiation as a function of frequency of valent and deformation vibrations (wavenumber, cm-1) for chemical bonds. We used the IR Fourier spectroscopy method (IR spectra with Fourier Transformation,) to investigated the grain composition in maize hybrids - ZP 341, ZP 434 and ZP 505 created at Maize Research Institute (Zemun Polje, Belgrade, Serbia). The resulted spectra differed in peak number and intensity, and in oscillation frequency. Particularly, there are 20 to 23 peaks and characteristic spectral bands within the wavenumber range of 400 to 4000 cm-1. Characteristic spectral bands were analyzed for each hybrid with regard to absorption intensity in %, experimentally determined wavenumber in cm-1, and published wavenumber range. A comparison of these peaks to reference IR spectra from databases revealed biogenic organic molecules: alcohols, amines, esters, alkanes, carboxylic acids, alkenes, aldehydes, ketones and esters in the studied grain hybrids. In a typical IR spectrum of maize hybrid ZP 341 there were three most distinct bands with wavenumbers of 3400, 2900 and 1000 cm-1. Four peaks (3400, 2950, 1700 and 1000 cm-1) were characteristic of the ZP 434 hybrid, and eight major peaks of 3400, 2900, 2850, 1750, 1700, 1450, 1150 and 1000 cm-1 were observed in ZP 505. That is, the grain charterisics in ZP 505 slightly differ from those in ZP 341 and ZP 434, whereas in ZP 341 and ZP 434 the grain structure is more similar. In general, Serbian hybrids are characterized by high quality, productivity and technological suitability. The developed methodology for IR spectra recording and analysis in grain allows to reveal the composition and structure of biogenic compounds. It is important not only for diagnosis and breeding, but also for the development of biotechnological screening methods, or the estimation of grain storage time.

A comparison of these peaks to reference IR spectra from databases revealed biogenic organic molecules: alcohols, amines, esters, alkanes, carboxylic acids, alkenes, aldehydes, ketones and esters in the studied grain hybrids. In a typical IR spectrum of maize hybrid ZP 341 there were three most distinct bands with wavenumbers of 3400, 2900 and 1000 cm-1. Four peaks (3400, 2950, 1700 and 1000 cm-1) were characteristic of the ZP 434 hybrid, and eight major peaks of 3400, 2900, 2850, 1750, 1700, 1450, 1150 and 1000 cm-1 were observed in ZP 505. That is, the grain charterisics in ZP 505 slightly differ from those in ZP 341 and ZP 434, whereas in ZP 341 and ZP 434 the grain structure is more similar. In general, Serbian hybrids are characterized by high quality, productivity and technological suitability. The developed methodology for IR spectra recording and analysis in grain allows to reveal the composition and structure of biogenic compounds. It is important not only for diagnosis and breeding, but also for the development of biotechnological screening methods, or the estimation of grain storage time.",

AB  - This paper discusses the application of the infrared (IR) spectroscopy method for determination of structural properties of maize hybrid grains. The IR spectrum of maize grain has been registered in the following hybrids: ZP 341, ZP 434 and ZP 505. Izvršeno je registrovanje infracrvenog spektra zrna hibrida kukuruza: ZP 341, ZP 434 i ZP 505. Pokazano je postojanje različitih spek tralnih traka, kako po broju i intenzitetu, tako i po njihovom obliku, frekvenciji i kinetici. AB  - In this paper the changes on growth, photosynthesis and water relations were tested by non-invasive fluorescence method. The applications of this method allow to determine some functional properties of prestigious maize inbred lines with erect top leaves. So the temperature dependency of delayed fluorescence intensity maximum of ZPPL 16 is observed at higher temperatures than for ZPPZ 62. This fact correlates with low values of phase transition of activation energy Ea in thylakoid membrane and accompanied by a decrease of the angle and area of the leaf, as well as with the content and the rate of water release from the seed. It seems reasonable to assume that, DF can be applied in breeding and maize hybrid seed production for the estimation of prestigious maize inbred lines and their resistance adaptability to increased and high temperatures, as well as, to drought.

Mechanisms and parameters of transients and oscillations of delayed chlorophyll fluorescence induction processes in the excited thylakoid membrane of the maize intact leaf

AB  - The intact leaves of four maize hybrids: ZP 360, ZP 46A, Zp 633 i ZP 704 and two maize inbred lines ZPR 70ž i Oh 43, tolerant and resistant to the impact of increased and high temperatures and drought were observed in the present study. The improved, non-invasive photosynthetic-fluorescence method was applied in studying of complex delayed chlorophyll fluorescence induction processes that were resolved into transients: A, B, C, D and E in which instabilities, fluctuations and oscillations can occur. The interrelationship between transients and oscillations of the delayed chlorophyll fluorescence induction processes occurred in the intact leaves of maize inbreds and hybrids that were grown in the green-house under conditions of different air drought, impacts of temperatures and the water deficit in the medium, when their thylakoid membranes had been excited and significant structural and functional changes had occurred. The existence of transients: A, B, C, D and E of delayed chlorophyll fluorescence induction processes was determined. The time and physico-chemical parameters of these transients were presented. The mechanisms of their generation were explained. The properties of oscillatory performances of delayed chlorophyll fluorescence induction processes were determined. The conditions under which instable state and fluctuations occurred and oscillations were established in the excited thylakoid membrane of the intact leaf of maize inbreds and hybrids were presented. The hypothesis on the mechanism of establishing of oscillations and their interrelation with transients of delayed chlorophyll fluorescence induction processes was presented.
AB  - Razloženi indukcioni procesi zakasnele fluorescencije (ZF) hlorofila na tranzijente: A, B, C, D i E nastaju kada se segment intaktnog lista linija i hibrida kukuruza drži u mračnoj komori fosforoskopa znatno kraće vreme (30 s ≤ τ ≤ 240 s), pre njegovog intermitentnog osvetljavanja sa belom svetlošću. Tranzijente indukcionih procesa: A, B, C, D i E karakterišu njihovi vremenski parametri: tA, tB, tC, tD i tE, promena intenziteta tranzijenata, parametri i mehanizmi njihovog nastajanja. Razloženi indukcioni procesi ZF hlorofila intaktnog lista linija i hibrida kukuruza na tranzijente: A, B, C, D i E praćeni su pojavom i različitim nivoima vrednosti energije aktivacije (Ea, kJ/mol) koje odgovaraju različitim kritičnim temperaturama faznih prelaza u pobuđenoj tilakoidnoj membrani. Mehanizmi nastajanja tranzijenata indukcionih procesa: A, B, C, D i E imaju dve prirode. Tranzijenti A i B su fizičke, a tranzijenti: C, D i E su hemijske prirode. Pokazano je da nastajanje tranzijenata: B, C, D i E istovremeno prati uspostavljanje oscilacija indukcionih procesa ZF hlorofila. Po našoj hipotezi, međuzavisnost tranzijenata i oscilacija indukcionih procesa ZF hlorofila dovode se u neraskidivu vezu. Oscilovanje indukcionih procesa ZF hlororfila objašnjava se strukturnim i funkcionalnim promenama u pobuđenoj tilakoidnoj membrani intaktnog lista linija i hibrida kukuruza koji su gajeni u uslovima vazdušne suše, povećanih temperatura i deficita vode u podlozi.
