Modelling of Genotype × Environment interaction for grain yield of late maturity maize hybrids in Serbia by climate variables

AB  - With global climate change, including unpredictable geographic and temporal weather patterns causing significant Genotype × Environment interaction (GEI), modelling by climate variables can reveal their influence on phenophases of maize (Zea mays L.) development. The objectives of this study were dissection of the phenotypic variation of grain yield of late maturity maize hybrids grown in multi-environment trial, and quantification of the influence of climatic factors on the GEI for each vegetative and reproductive phenophase. Eight FAO 700 maize hybrids were evaluated across five locations in Serbia during 2020 and 2021 years. The hierarchy of sources of variation according to three-way ANOVA were: Year (Y) > Location (L) > Location×Genotype (G) > L×Y×G > G > L×Y > Y×G. The average maximum temperature (mxt, 22.1%), average minimum temperature (mnt, 19.2%), average mean temperature (mt, 18.2%) and relative humidity (rh, 15.1%) in April significantly influenced emergence stage. The mxt (21.1%) and mt (15.7%) in May influenced significantly vegetative phases V1-V9. June contributed the largest percentage of the sum of squares of the GEI with mxt (25.2%), mnt (20.9%), mt (16.1%) influencing vegetative phases V10-V18 and tassel emergence. In July mxt (17%), mt (15.6%), precipitation sum (15.2%), and sunshine hours sum (15.1%), influenced R1, R2, R3, and R4 reproductive phases. In August mxt (23.2%), mnt (20.8%), mt (15.7%), rh (17.1%) influenced R5 reproductive phase. The extreme heat as a stressor had a more critical role for late maturity maize hybrids production than drought in crucial phenophases of maize development.

AB  - The study was carried out at the Maize Research Institute “Zemun Polje”, Belgrade, Serbia during 2021
growing season in three location. Six inbred lines were crossed according to complete diallel method. In this
way 15 hybrids and 15 reciprocal combinations were obtained. Hybrid and reciprocal combinations derived
from these parental components were used in this paper.
The present study was carried out to study the variability, broad-sense heritability (H2
bs), genetic advance
(GA), correlation among traits, genotypic and phenotypic path analysis among grain yield and its components.
The results indicated that the genotypes were significantly different for all traits. For all traits, the phenotypic
coefficient of variation was higher than the genotypic coefficient of variation. The highest heritability was
found in 1000 kernel weight, followed by grain yield, ear diameter and anthesis-silking interval (ASI). In our
study Genetic Advance over Mean (GAM) for the traits ranged from lowest value (9.739%) for cob diameter to
the highest value (90.579%) for anthesis-silking interval.
According to results of path analysis, the trait 1000 kernel weight with the value of 0.365 has the strongest
direct positive effect on grain yield. Positive direct effects on grain yield were also observed for ear length
(0.202), ear diameter (0.248) and number of rows per ear (0.076), while negative direct effects were observed
for cob diameter (-0.057). The trait 1000 kernel weight had the highest indirect positive effects on grain yield
via ear diameter (0.232). The cob diameter had highest negative indirect effect on grain yield via ear diameter
(-0.048).
Coefficient of multiple determinations (R2
) had a value of 0.428. This indicated the important role of
additive gene effect in the inheritance of these traits and could be improved through simple selection.

worldwide agriculture with highest production and productivity. Maize is also an important
grain crop grown in Serbia, used as a component of feed. Many studies have been conducted
on correlations, heritability and path analysis on grain yield. The results have been widely
used in maize breeding programs. The present research was carried out at the Maize Research
Institute “Zemun Polje” in Serbia during the 2021 growing season effects of grain yield and
quantitative traits of maize. Six inbred lines were crossed according to complete diallel
method. In this way fifteen hybrids and fifteen reciprocal combinations were obtained. The

AB  - Methods of absorption and Raman spectroscopy were applied in this study to analyze functions and amounts of photosynthetic pigments in the leaf of maize inbred lines (M1-3-3Sdms, ZPPL186, and ZPPL225). Absorption spectroscopy was used to determine the dynamics of biosynthesis of chlorophyll a, chlorophyll b, and carotenoids. Raman spectroscopy was used to present the dynamics of biosynthesis of carotenoids and other compounds in terms of origin and kinetic form of the formation of spectra with all spectral bands. Organic molecules and compounds causing the formation of certain spectral bands in the Raman spectrum (carotenoids, phosphates, glycogen, amid III, and others) were determined. Conformational and functional changes of photosynthetic pigments in the leaves of the maize inbred lines, which occur due to changes in the ratio (quotient), were analyzed. The ratios (quotients) obtained indicate different contributions of valence oscillations of their chemical bonds, which inevitably alter the conformation of molecules and compounds. The results presented for the overall study point to minor biogenic differences among the maize inbred lines under study.
AB  - У овом раду примењене су методе за апсорпциону и Раманову спектроскопију у изучавању функција и количина фотосинтетских пигмената у листу инбред линија кукуруза: M1-3-3Sdms, ZPPL186 и ZPPL225. Помоћу апсорпционе спектроскопије утврђена је динамика биосинтезе свих форми хлорофила: хлорофил a, хлорофил b, хлорофили a + b и каротеноида. Помоћу Раманове спектроскопије показана је динамика биосинтезе каротеноида и других једињења у погледу места и кинетичке форме настајања спектара са свим спектралним тракама. Утврђени су органски молекули и једињења који условљавају настајање појединих спектралних трака у Рамановом спектру (каротеноиди, фосфати, гликоген, амид III и други). Анализиране су структурне и функционалне промене фотосинтетских пигмената у листу инбред линија кукуруза до којих долази због промене односа (количника). Добијени односи (количници) указују на различите уделе валентних осцилација и њихових хемијских веза, што неминовно мења конформацију молекула и једињења. Изложени резултати указују на мање биогене разлике између проучаваних инбред линија кукуруза.

AB  - The history of maize breeding in Maize Research Institute Zemun Polje has
been going on for more than seven decades. Maize breeders in the Institute
create their breeding programs in such way to respond to actual market
demands and modern trends in maize cultivation but also by using available
new technologies that accelerate and increase the efficiency of maize hybrid
development. One of the tools that makes this possible is double haploid (DH)
breeding technology, which has been applied in the Maize Institute since 2014.
Breeding programs with the use of DH are realized by the use of inducers with
high induction rate, additional winter generations and the development of a
reliable system (markers) induced haploid recognition. In this way, ZP 4019
was created at the Institute Zemun Polje as the first official maize hybrid with
a parental inbred line obtained by DH technology. This is a modern maize
hybrid intended to respond to the changes in maize cultivation in recent years
caused by increasingly frequent periods of draught, unfavorable precipitations
and temperatures in the flowering and grain filling stages, as well as demands
for higher yields and easy grain harvesting without the need for grain drying.
ZP 4019 plant habitus is characterized by a low stem, a well-developed root
system, a significant leaf area, erect position of the upper leaves and a lower
planted ear which enables growing in higher densities. In testing on different
agroecological environments, this hybrid showed high adaptability and
stability. ZP 4019 was tested in official test trials of the Section for plant variety
registration during 2021 and 2022 and performed significantly better than
check hybrids NK PAKO and ZEROS, while its grain moisture content was
below the official threshold for the FAO 400 maturity group.

AB  - Maize breeding program at the Maize Research Institute "Zemun Polje" begun
during the fifties of the last century and it was based on the principles of
inbreeding hybridization. During the sixties, first maize hybrids were
registered and each subsequent decade a new generation of hybrids was
created. So far, more than 800 maize hybrids have been registered in Serbia
and more than 170 hybrids abroad. Testing program includes field evaluations
of experimental hybrids in a large number of locations in Serbia, paying
attention to the correct choice of environments. Each season 5000-7000 new
hybrid combinations have been tested in Serbia. Additionally, the most
promising hybrids of different maturity groups are being tested at more than
20 locations abroad, with the aim of selecting the best hybrids for each foreign
market. In order to accelerate breeding procedure, service of winter generation
in Chile is used, with the aim of obtaining two generations during one calendar
year. Since 2014, the application of double haploid technology in maize
breeding has started. So far, 13000 double haploid lines have been created from
350 original populations. Optimization of each step in the methodology
towards the higher efficiency is the main task for breeders: higher induction
rate, higher duplication rate, better agronomic traits of inducers. This year the
first hybrid containing a double haploid line has been registered. In addition,
several hybrids developed using this method are in registration procedure.
Application of molecular markers represents an essential part of modern
breeding programs. Use of SNP markers became a routine tool in breeding
program of Maize Research Institute. In the last few years, 400 most elite ZP
maize inbred lines were characterized using this type of markers. In addition
to the creation of hybrids with standard grain quality, breeding program of
specific maize hybrids exists and it includes breeding for hybrids with white
endosperm, sweet maize and popcorn.
Breeding for herbicide tolerance resulted in registration of several cycloxidim
tolerant hybrids during last decade. Also, several hybrids with red grain color,

which are characterized by higher anthocyanin content, have been registered
so far. Recent innovation in maize breeding program is implementation of
cloud-based software platform, which will enable better data collection and
management, creation of new experiements and improved statistical analysis.

AB  - Sastav zrna kukuruza, posebno skroba, proteina i lipida, ključan je za ishranu
ljudi i životinja. U savremenim programima oplemenjivanja kukuruza prioritet se
daje prinosu, što dovodi do smanjenja sadržaja proteina i povećanja sadržaja skroba.
U potrazi za zdravijom hranom, lokalne populacije privlače pažnju kao vredan
genetički resurs. Cilj ovog rada je da se identifikuju lokalne populacije kukuruza
koje su, kada su ukrštane sa divergentnim tester linijama, proizvele potomstvo sa
povećanim sadržajem proteina, skroba ili lipida uz održavanje zadovoljavajućeg
prinosa. Trideset i jedna lokalna populacija kukuruza je ukrštena sa tri genetički
različite linije testera. Nutritivne vrednosti dobijenih test cross hibrida procenjene
su tokom dve godine i na sedam lokacija korišćenjem bliske infracrvene (NIR)
spektroskopije za nedestruktivnu i brzu analizu. Istraživanja su otkrila značajan
uticaj sadržaja skroba na prinos hibrida, uz obrnutu korelaciju između sadržaja
proteina i skroba. Populacije kukuruza sa visokim sadržajem skroba dale su hibride
sa niskim sadržajem proteina i obrnuto. Kod sadržaj lipida u zrnu nisu utvrđene
jasne zakonitosti. PCA analiza je identifikovala 12 populacija sa visokim sadržajem
proteina, 12 sa visokim sadržajem skroba i 7 sa visokim sadržajem lipida. Lokalne
populacije kao što su MB13 i MB197, identifikovane su kao donori osobina
važnih za prinos zrna i sadržaj proteina (vrednosti OKS od 0,51* (9,96%) i 0,57*
(9,89%) respektivno). Populacija MB594 je pokazala da unosi visok sadržaj skroba
(70,39%), dok je MB632 pokazala potencijal za poboljšanje sadržaja lipida (4,39%)
i visoko značajnu pozitivnu OKS vrednost od 0,33**. Rezultati rada su pokazali da
NIR spektroskopija kao nedestruktivna i brza metoda može poslužiti za procenu
sastava zrna kukuruza i da identifikuje lokalne populacije kukuruza za programe
oplemenjivanja usmerenih na poboljšanje specifičnih nutritivnih osobina.

is crucial for both human and animal nutrition. Modern maize breeding programs
often prioritize yield, leading to a decrease in protein content and an increase in
starch content. In search of healthier food options, maize landraces have gained
attention as a valuable genetic resource. This study aimed to identify maize landraces
that, when crossed with divergent tester lines, produced progeny with enhanced
protein, starch, or lipid content while maintaining satisfactory yield. Thirty-one
local maize populations were crossed with three genetically diverse tester lines.
The nutritional values of resulting test cross hybrids were assessed over two years
and across seven locations using near-infrared (NIR) spectroscopy for non-destructive
and rapid analysis. The study revealed significant impact of starch content
on hybrid yield, with an inverse correlation between protein and starch content.
Landraces with high starch content produced hybrids with low protein content
and vice versa. Lipid content showed no clear pattern. PCA analysis identified
12 landraces with high protein content, 12 with high starch content, and 7 with
high lipid content. Certain landraces, such as MB13 and MB197, were identified
as donors of traits important for grain yield and protein content (GCA values of
0.51* (9.96%) and 0.57* (9.89%) respectively). Landrace MB594 exhibited a high
starch content (70.39%), while MB632 showed potential for lipid content (4.39%)
improvement and a highly significant positive GCA value of 0.33**. This study demonstrates
the utility of NIR spectroscopy as a non-destructive and rapid method
for assessing maize grain composition. It identified promising maize landraces for
breeding programs focused on enhancing specific nutritional traits.

FAO 500 predstavljaju odličnu kombinaciju visokog prinosa i nižeg sadržaja vlage
u zrnu. Mogu se ubirati i u klipu i u zrnu, što ih čini veoma dobrom alternativom
za hibride kasnih grupa zrenja koji se odlikuju visokim potencijalom za prinos
ali i visokim sadržajem vlage u žetvi. Hibrid ZP 5557 registrovan je 2023. godine.
U ogledima Sortne komisije je ispitivan u periodu 2021-2022. godine. Karakteristično
je da su obe godine bile nepovoljne za gajenje kukuruza, što se naročito
odnosi na 2022. godinu. Hibrid ZP 5557 je u obe godine ispitivanja ostvario viši
prosečan prinos u odnosu na prosek standardnih hibrida, uz niži sadržaj vlage i
manji procenat poleglih i slomljenih biljaka. U dvogodišnjem proseku ostvario je
prinos od 104,4% u odnosu na prosek standardnih hibrida. Na osnovu rezultata
postignutih u ogledima sortne komisije, hibrid ZP 5557 biće uključen u postkomisijske
oglede Instituta za kukuruz i u slučaju da ponovi dobar rezultat biće
započeta komercijalizacija ovog hibrida.

AB  - Plant genetic resources of any country represent its natural wealth. Gene Bank
of the Maize Research Institute "Zemun Polje" with over 5,000 samples
represents national guardian of such. Local maize populations, tributing to their
diversity, represent a key source of desirable traits for breeding programs, such
as nutritional values for protein, starch and lipids. Today’s corn hybrids are
based on a narrow genetic base, where in race for yield, often insufficient
attention is paid to the nutritional composition of the hybrids themselves.
Correct selection of material from the Gene Bank can only be achieved with
pre-breeding selection. Such prebreeding would cover wide material and
should detect and reduce it to a desirable and more acceptable volume that can
further be introduced into commercial breeding programs. This paper covered
analysis of protein, starch and lipid content in grain of 31 local maize
populations, crossed with two commercial testers ZPL-217 (BSSS) and ZPL-
255/75-5 (Lancaster Sure Crop) with line x tester method used for combinig
ability calculation. According to the results the best combiner for higher lipid
content is A3 landrace with a GCA value of 0.33*, for protein content landraces
A9 and A 38 with GCA values of 0.51* and 0.57*, respectively, were found.
As for populations A 17 and A 37 with the GCA values of 1.52* and 1.36*,
respectively, they proved to be the best combiners for starch. Mentioned
populations showed significant GCA values with a probability level of 0.05,
showing their possible significance in further breeding application.

AB  - Genetska konstitucija, uslovi gajenja, uključujući agrotehniku i manipulaciju
posle žetve, utiču na kvalitet zrna kukuruza. Zahtevi za kvalitet se razlikuju od
jedne do druge upotrebe i uvek moraju biti definisani u skladu sa metodama koje
se primenjuju u preradi i korišćenju. Ciljevi ovog istraživanja bili su da se : i) procene prinos, komponente prinosa i kvaliteta zrna šest elitnih hibrida kukuruza,
ispitivanih tokom 5 godina u mikro i makro ogledima; ii) utvrde međusobni odnosi
ispitivanih osobina u zavisnosti od efekta hibrida, godine i roditeljskih komponti.
Efekat godine je u velikoj meri uticao na varijaciju svih ispitivanih parametara,
efekat oca nije bio statistički značajan za komponente prinosa, dok su efekti i oca i
majke bili značajni za većinu ispitivanih osobina kvaliteta. PCA analiza je ukazala
na značajan uticaj genetske konstitucije hibrida na fizičke parametre i strukturu
zrna. Hibridi oca LM3 imali su veću apsolutnu masu, udeo mekog endosperma
i otpornost na mlevenje, a niži indeks apsorpcije vode i udeo perikarpa. Hibridi
majke LF2 su imali veći udeo endosperma, dok su hibridi majke LF1 imali veći udeo
klice i ulja u zrnu. Rezultati su ukazali da je moguće simultano oplemenjivanje na
povećanje skroba i proteina (uz povećanje prinosa), kao i oplemenjivanje na povećanje sadržaja proteina i ulja (uz smanjenje prinosa). Istovremeno oplemenjivanje
u pravcu povećanja sadržaja skroba i ulja bilo bi otežano višestrukim negativnim
odnosima ove dve osobine. Međutim, iako su utvrđene statistički značajne razlike
u parametrima kvaliteta, one nisu napravile kvalitativnu razliku u proizvodnim
ogledima. Zbog toga je potrebno formirati posebne programe oplemenjivanja za
različite namene.
AB  - Genetic background, environmental conditions, agricultural practices and
post-harvesting influence maize grain quality. The quality requirements differ
from one use to another, and need to be defined in accordance with the methods
applied in processing and utilisation. The objectives of this research were: i) to
evaluate yield and grain quality traits of six elite maize hybrids, tested during 5-year
micro and macro trials; ii) to determine the evaluated traits relationshipaccording
to hybrid-, year- and parental components-dependence pattern. The year effect
greatly influenced the variation of all examined parameters, the male effect was not
significant for yield components, while the effects of both, male and female parent
were significant for most of the examined quality traits. PC analysis indicated a
significant effect of the hybrid’s genetic composition on physical parameters and
kernel structure. Hybrids of male LM3 had higher 1000 kernelweight, portion of
soft endosperm and resistance to grinding, and lower water absorption index and
pericarp portion. Hybrids of female LF2 had a higher portion of endosperm, while
hybrids of female LF1 had higher portion of germ and oil in the grain. Accordingly,
simultaneous breeding for increased starch and protein resulted in yield increase,
forincrease protein and oil content in yield reduction, while simultaneous breeding
for increased starch and oil content is hampered by multiple negative relationships.
Although significant differences in quality parameters were determined, they did
not make a qualitative difference in production trials. Therefore, it is necessary to
form special breeding programmes for different purposes.

The interval of variation for standard deviation of average ranks for tested maize hybrids
was from 1.3 (ZP4, Ch) to 2 (ZP2) indicating satisfying accordance of utilized parametric
and non-parametric stability measures. The highest average grain yield across all tested
environments was observed for ZP1, which was second most stable hybrid. The most
stable late maturity maize hybrid tested over ten environments was proven to be ZP6 with
sum of ranks and average rank of 37 and 1.8, respectively, with standard deviation of
average rank 1.5. AB  - Već više od sedam decenija Institut za kukuruz "Zemun Polje" kontinuiranim radom na razvoju i stvaranju novih hibrida kukuruza prati korak sa savremenim trendovima u oplemenjivanju kukuruza. Oplemenjivači u Institutu za kukuruz “Zemun Polje” svojim radom na stvaranju novih hibrida kukuruza nastoje da odgovore na aktuelne potrebe tržišta ali i da primenom savremenih tehnologija (dihaploidi, molekularni markeri...) ubrzaju i povećaju preciznost i efikasnost svojih programa. Uvođenjem i ispitivanjem genetičkog materijala različitog porekla (ex-pvp materijal) u selekcionim programima se povećava diverzitet oplemenjivačkog materijala. lako se, zbog klimatskih promena, tržište gajenja kukuruza u Srbiji pomera ka srednje ranim grupama zrenja, na tržištu postoji i dalje potreba za savremenim, prinosnim i stabilnim hibridima kasnijih grupa zrenja. Iz tog razloga Institut za kukuruz “Zemun Polje” posvećuje značajnu pažnju u svojim programima razvoju linija i hibrida kukuruza FAO 600-700 grupa zrenja. Kao rezultat tih programa je nastao i hibrid ZP 6090 koji je ispitivan u ogledima Sortne komisije Republike Srbije tokom 2021. i 2022. godine. Tokom obe godine ispitivanja na ukupno 13 lokacija, hibrid ZP 6090 se istakao odlicnim performansama. Hibrid ZP 6090 je hibrid namenjen primeni savremenih tehnologija gajenja kukuruza. Ovaj hibrid ima modernu arhitekturu bilike koju karakteriše umerena visina, nizak polozaj klipa i uspravan položaj listova. Na ovaj način omogućeno je gajenje u većim gustinama u odnosu na druge hibride iste grupe zrenja kao i primena intezivnog načina gajenja. Hibrid ZP 6090 pokazuje izraženu otpornost prema poleganju, a pokazuje i značajnu tolerantnost na sve ekonomski važnije bolesti i štetocine kukuruza. Zbog velike lisne površine, kvalitetnog zrna i izraženog staygreen svojstva pogodan je i za silažnu upotrebu.

AB  - Institut za kukuruz "Zemun Polje" (MRIZP) razvija program oplemenjivanja kukuruza vise od 75 godina. Stvaranje novih hibrida, koji po svojim najvažnijim agronomskim osobinama prevazilaze hibride prethodne generacije predstavlja primarni zadatak svakog oplemenjivača kukuruza. Prisustvo ZP hibrida kukuruza na tržištima Republike Srbije, ali i u inostranstvu, zahteva stvaranje hibrida za različite agroklimatske uslove, kao i za razlicite namene. Spajanje različitih naučno istraživačkih disciplina dovelo je do ubrzanja i povećanja tačnosti procesa oplemenjivanja kukuruza, a samim tim i do smanjenja trajanja ciklusa oplemenjivanja kukuruza u svetu do pet, a u MRIZP-u na 6-7 godina. Od 2014. godine MRIZP uspešno primenjuje program dvostrukih haploida (DH) u oplemenjivanju. Primenom takvog pristupa storeno je više od 13.000 novih DH inbred linija. Kao rezultat primene DH tehnologije, registrovan je prvi hibrid ZP 4019. MRIZP intenzivno radi na karakterizaciji najelitnijeg materijala uz pomoc SNP (Single Nucleotide Markers) markera koristeći 25k SNP Ilumina Infinium Array. Na ovaj način je ostvaren najprecizniji uvid u germplazmu oplemenjivača, odnosno odredjivanjem genetičke udaljenosti (GD). Uz pomoć GD vrednosti povećana je verovatnoća dobijanja superiornih inbred linija i hibrida kukuruza. Već dve sezone MRIZP je započeo visokoinformativnu fenotipizaciju u polju (HTFP), koristeći RGB i multispektralne kamere zasnovane na dronovima za ekstrakciju i vizuelizaciju podataka po parceli. U ovoi fazi istražujemo mogućnost zamene ručnih merenja (kao što je broj i poleganje biljaka), dok je naš konačni cilj da upotrebimo HTFP za donošenje presudnih odluka za hibrid u našim programima oplemenjivanja.

AB  - In vivo metoda dupliciranih haploida u Institutu za kukuruz „Zemun Polje“ koristi se u oplemenjivanju kukuruza od 2014. godine. Pristup omogućava dobijanje potpuno homozigotnih inbred linija tzv. duplohaploidnih (DH) linija za samo dve godine (dve do tri generacije), za razliku od konvencionalnog oplemenjivanja gde je potrebno bar sedam do osam generacija kako bi se postigla neophodna homozigotnost. Metoda se sastoji iz nekoliko koraka: 1) indukcija haploida iz odabranih populacija ukrštanjem sa linijom induktorom haploida (prva godina/letnja generacija); 2) selekcija haploidnih zrna (prva godina/letnja generacija); 3) duplikacija hromozoma (druga godina/zimska generacija) i 4) umnožavanje dobijenih DH linija (druga godina/letnja generacija). Do sada je dobijeno oko 13000 DH linija iz oko 350 populacija koje pripadaju različitim heterotičnim grupama. U okviru programa sprovode se i aktivnosti sa ciljem razvoja novih induktora haploida sa povišenim procentom indukcije haploida i poboljšanim agronomskim svojstvima. Osam induktora-kandidata je specifičnom procesom selekcije odabrano i u 2023. godini ocenjeno u mini ogledu sa komercijalno dostupnim induktorima, a mereni su visina biljke, produkcija polena, ozrnjenost u ukrštanju, kao i procenat indukcije haploida. U okviru rada na optimizaciji metode, ispitivan je i uticaj populacije (heterotične grupe) i spoljašnje sredine na uspešnost metode. Utvrđen je značajan uticaj spoljašnjih uslova, kao i heterotične grupe odnosno genotipa na indukciju i duplikaciju, kao i konačnu uspešnost metode. Prvi hibridi sa DH linijom kao roditeljskom komponentom već su registrovani od strane Sortne komisije Republike Srbije za komercijalno gajenje u Srbiji.

abstract = "In vivo metoda dupliciranih haploida u Institutu za kukuruz „Zemun Polje“ koristi se u oplemenjivanju kukuruza od 2014. godine. Pristup omogućava dobijanje potpuno homozigotnih inbred linija tzv. duplohaploidnih (DH) linija za samo dve godine (dve do tri generacije), za razliku od konvencionalnog oplemenjivanja gde je potrebno bar sedam do osam generacija kako bi se postigla neophodna homozigotnost. Metoda se sastoji iz nekoliko koraka: 1) indukcija haploida iz odabranih populacija ukrštanjem sa linijom induktorom haploida (prva godina/letnja generacija); 2) selekcija haploidnih zrna (prva godina/letnja generacija); 3) duplikacija hromozoma (druga godina/zimska generacija) i 4) umnožavanje dobijenih DH linija (druga godina/letnja generacija). Do sada je dobijeno oko 13000 DH linija iz oko 350 populacija koje pripadaju različitim heterotičnim grupama. U okviru programa sprovode se i aktivnosti sa ciljem razvoja novih induktora haploida sa povišenim procentom indukcije haploida i poboljšanim agronomskim svojstvima. Osam induktora-kandidata je specifičnom procesom selekcije odabrano i u 2023. godini ocenjeno u mini ogledu sa komercijalno dostupnim induktorima, a mereni su visina biljke, produkcija polena, ozrnjenost u ukrštanju, kao i procenat indukcije haploida. U okviru rada na optimizaciji metode, ispitivan je i uticaj populacije (heterotične grupe) i spoljašnje sredine na uspešnost metode. Utvrđen je značajan uticaj spoljašnjih uslova, kao i heterotične grupe odnosno genotipa na indukciju i duplikaciju, kao i konačnu uspešnost metode. Prvi hibridi sa DH linijom kao roditeljskom komponentom već su registrovani od strane Sortne komisije Republike Srbije za komercijalno gajenje u Srbiji.",

AB  - Инфракрасная (ИК) спектроскопия и спектроскопия комбинационного рассеяния (КР) — современные методы, на основе которых успешно формируются биотехнологические подходы, поз-воляющие проводить генетический и функциональный анализ отдельных органов и тканей растений на молекулярном уровне. В представленной работе с помощью спектроскопии поверхностного внут-реннего отражения (ПВО), которая представляет собой модификацию ИК-спектроскопии, впервые зарегистрированы и выявлены различия в содержании и конформации биомолекул в гомогенатах целого зерна гибридов кукурузы ZP 735 и его компонентов (эндосперм, перикарп, зародыш). Нашей целью была разработка методологии спектроскопии поверхностного внутреннего отражения для идентификации органических молекул, их содержания и конформации в семени кукурузы и его тканях. Объектом исследования служили зерна гибрида кукурузы (Zea mays L.) ZP 735 (оригина-тор — Maize Research Institute, Zemun Polje, г. Белград, Сербия). Методом случайной выборки было отобрано 30 зерен. Для получения ПВО-спектров образцы целых зерен и их частей (эндо-сперма, перикарпа и зародыша) измельчали, гомогенизировали и помещали в специальную ячейку прибора. Для регистрации ПВО-спектров использовали спектрометр (Nicolet™ iS20 FTIR Spectro-meter, «Thermo Scientific», США) с алмазом (diamond ATR crystal, «Thermo Scientific», США), для анализа спектров — пакет программ («Thermo Scientific», США). ПВО-спектры сопоставляли в библиотеке данных программы Advanced ATR correction Algorithm. Метод ПВО-спектроскопии ос-нован на отражении пучка света на границе раздела двух фаз: фазы кристалла с высоким показа-телем преломления, входящего в состав прибора ПВО, и фазы исследуемого образца с более низ-ким показателем преломления. В процессе измерения луч света проникает на небольшую глубину в фазу образца, где частично поглощается. При последующих облучениях образца это явление повторяется, и в результате регистрируется ПВО-спектр. Доказано, что в диапазоне от 400 см1 до 4000 см1 ПВО-спектры целого зерна, эндосперма, перикарпа и зародыша гибрида ZP 735 были аналогичны полученным ранее ИК-спектрам зерна, что свидетельствует о возможности использо-вания нового метода для технологий молекулярной селекции. Расположение полос ПВО-спектра характеризует различные формы колебаний валентных связей функциональных групп органических молекул семени, что позволяет идентифицировать не только наличие определенных молекул, но и их конформацию. Доказано, что с помощью анализа амплитуды полос ПВО-спектра (максималь-ная амплитуда интенсивности и высокая амплитуда интенсивности) в тканях семени можно кон-тролировать изменение содержания ряда органических соединений: протеинов, липидов, сахаров, сложных эфиров, амидов, кетонов, альдегидов, карбоновых кислот, простых эфиров, фенолов, спиртов, ароматических углеводородов, ациклических соединений, алкенов, алканов и алкинов. Полученные результаты важны для тестирования наличия в зерне необходимых органических со-единений или изменения их конформации в процессе селекции. К важным преимуществам метода ПВО-пектроскопии по сравнению с ИК-спектроскопией относятся, с одной стороны, сохранение нативности объекта (возможность проведения исследования без фиксации или окрашивания) и простая пробоподготовка, с другой — эффективная оценка содержания и конформации молекул с высокой чувствительностью (разрешение порядка 1,0 см1). Реализация разработанной методики для формирования технологии молекулярной селекции повысит рентабельность культивирования и эффективность селекции не только кукурузы, но и других сельскохозяйственных растений.
AB  - Infrared (IR) spectroscopy and Raman spectroscopy (RS) are modern methods on the basis of which biotechnological approaches are being successfully developed that allow genetic and func-tional analysis of individual plant organs and tissues at the molecular level. In the present work, using surface internal reflection spectroscopy (SIR), which is a modification of IR spectroscopy, differences in the content and conformation of biomolecules in grain homogenates and its components (endo-sperm, pericarp, germ) of ZP 735 maize hybrids were first recorded and revealed. Our goal was to develop a methodology for surface SIR for the identification of organic molecules, their content and conformation in corn seed and its tissues. The grains of the corn hybrid (Zea mays L.) ZP 735 (origi-nator Maize Research Institute, Zemun Polje, Belgrade, Serbia) served as the object of the study. Thirty grains were selected by random sampling. To obtain SIR spectra, samples of grains and their parts (endosperm, pericarp, and embryo) were crushed, homogenized, and placed in a special cell of the device. A spectrometer (FT-IR spectrophotometer, Thermo Scientific, USA) with diamond (dia-mond ATR crystal, Thermo Scientific, USA) was used to record the SIR spectra, and a software package (Thermo Scientific™ ) was used to analyze the spectra. The SIR spectra were compared in the data library of the Advanced ATR correction Algorithm program. The SIR spectroscopy method is based on the reflection of a light beam at the interface between two phases: the phase of a crystal with a high refractive index, which is part of the SIR device, and the phase of the sample under study with a lower refractive index. During the measurement, the light beam penetrates to a small depth into the phase of the sample, where it is partially absorbed. During subsequent irradiation of the sample, this phenomenon is repeated, and as a result, the SIR spectrum is recorded. It has been proven that in the range from 400 cm-1 to 4000 cm-1 ATR spectra of whole grain, endosperm, pericarp and embryo of the ZP 735 hybrid are similar to the previously obtained IR spectra of grain, which indicates the possibility of using a new method for molecular breeding technologies. The location of the bands of the SIR spectrum characterizes various forms of vibrations of the valence bonds of the functional groups of the organic molecules of the seed, which makes it possible to identify not only the presence of certain molecules, but also their conformation. It has been proved that by analyzing the amplitude of the SIR spectrum bands (maximum intensity amplitude and high intensity amplitude), it is possible to control changes in the content of a number of organic compounds in seed tissues: proteins, lipids, sugars, esters, amides, ketones, aldehydes, carboxylic acids, simple ethers, phenols, alcohols, aromatic hydrocarbons, acyclic compounds, alkenes, alkanes and alkynes. The obtained results are important for testing the presence of the necessary organic compounds in the grain or changes in their confor-mation during the selection process. The important advantages of SIR spectroscopy compared to IR spectroscopy include, on the one hand, the preservation of the nativeness of the object (the possibility of conducting research without fixing or staining the object) and simple sample preparation, on the other hand, an effective assessment of the content and conformation of molecules with high sensitivity (resolution about 1.0 cm-1). The implementation of the developed methodology for the formation of molecular breeding technology will increase the profitability of cultivation and the efficiency of breed-ing not only corn, but also other agricultural plants.

Grain Yield Predictions of ZP Maize Hybrids

AB  - Maize hybrids are created by crossing inbreds of
opposite heterotic groups, where the discovery of
genetic distances (GD) is of great significance. With
the presence of modern breeding tools for discovering
heterotic pairs and GDs in used material an efficiency in
breeding can be increased for more precisely planned
field work.
For this study 11 crosses were made in order to
compare the prediction of data obtained by GDs
between crossed inbreds with the results of same
hybrids from field testing. Crosses were done in 2020
according to the GD suggestions which ranged from
0.386 to 0.465 to check out is it possible to predict
specific superior hybrid performance before the field
results itself. 25k SNP Ilumina Infinium Array for maize
was used to determine GD values.
As a check ZP 457-market accepted hybrid (registered
in 2018 in Republic of Serbia) was used together with
10 other hybrids. All 11 hybrids shared the same female
component (Lancaster Sure Crop heterotic group),
while male components from opposite heterotic groups
(BSSS and Iowa dent) were used. GD values of used
males ranged from 0.190 to 0.412 clearly stating one
broad heterotic group. The most famous heterotic pair
B-73 x Mo-17 as well as two Institute’s top selling
hybrids (ZP 434 and ZP 606) were also included for
GD comparison with test hybrids with the GD values of
0.488, 0.466 and 0.462 respectively.
Review of B-73 x Mo-17 through ZP 434 and ZP 606
to the newly created hybrids suggests GD values are
decreasing with every new cycle of selection between
crossed material. Results from field testing compared
to GD values show somewhat positive correlation as
for ZP 3 exp which ranked 2nd highest GD value and
3rd rank in grain yield outyielding ZP 457 significantly,
but ZP 457 GD value of 0.458 (ranking third in GD)
suggests higher yielder should be though to find. Grain
yield results proved same hybrid ZP 457 to be one
of the lowest yielders being superior only to ZP 4123
despite high GD value. In top three yielders GD values
were in great correspondence for only one mentioned
combination ZP 3 exp, but for the ZP 4 exp and ZP
457 taking first and third position according to the GD
rank proved to be good only for 5th and 10th rank
within field results, while the lowest yielder ZP 4123
took 4th position according to GD values, having very
high value of 0.457.
This result proves SNP to be powerful tool in predicting
herotic pairs in maize and must be considered for
more precise planned field work. Even though GD
values being so informative on one side they do not
specifically imply to the solution for detecting the most
superior hybrid combination therefore leaving field
observations to confirm start assumptions.

AB  - Maize (Zea mays L.) is the third most grown cereal crop in the world, after wheat and rice. Immature dehusked maize ears harvested before fertilization, within 2-4 days after white silk emergence are classified as baby corn. The objective of this study was to investigate the suitability of baby corn cobs obtained from different maize hybrids for their enrichment with anthocyanins extracted from the black soybean seed coats. Three different maize hybrids, namely, a popcorn hybrid ZP 6119k, a sweet hybrid ZP 553su, a semi-flint with a short vegetation period (105 days) ZP 161, as well as black soybean genotype “Black Tokyo”, were used in this study. Plant material was grown in the experimental field of the Maize Research Institute at the location of Zemun Polje, Serbia in 2021. Maize was harvested after the emergence of silk from the ears (silking stage), while soybean plants were harvested in the stage of full maturity (R8). The baby maize cobs differed in length (9.5-20.0 cm) and girth (12-21 mm), the number of kernel rows (12-20), color, and length of the silk outside the husk (3-7 cm). Soybean seeds were manually dehulled and seed coat was ground in a laboratory mill prior to the extraction of anthocyanins. After dehusking and measuring, the baby corn cobs were washed and blanched for two minutes in boiling water, and then cooled. Subsequently, the baby corn cobs were immersed in a marinating brine enriched with anthocyanins extracted from the seed coat of black soybean, pasteurized, and kept in closed glass jars in a cool dry place for 30 days. After chemical and sensory analyzes, the product obtained from popcorn hybrid ZP 6119k was chosen as the most suitable for the preparation of the marinated baby corn product enriched with anthocyanins from black soybean

Optimization of the doubled haploid technology for temperate maize breeding programs: a case study from Maize Research Institute Zemun Polje

AB  - Doubled haploid (DH) technology is one of the several technological advances greatly impacting modern maize breeding. The main benefit for breeders is the development of parental inbred lines in one year comparing to four using conventional breeding. Maize Research Institute Zemun Polje (MRIZP) initiated doubled haploid program in maize in 2014. In six years up to 8.000 DH lines have been developed from close to 160 donor germplasm sources of different genetic backgrounds. During this period the research group engaged in the DH program identified several issues that need to be addressed in order to optimize the method for use in temperate regions including poor agronomic performance of haploid inducing lines, donor germplasm effect on the overall efficiency of DH method in maize breeding and selection strategies for improving test-cross performance of DH lines. To meet the challenges, MRIZP DH research group started a breeding program aiming at developing inducers with high and stable HIR adapted to temperate climate conditions. Two main goals were defined: development of haploid inducers with HIR up to 20% in a completely inducer genetic background, and second, to develop haploid inducers in 75 and 50% inducer genetic background with moderate HIR but better agronomic performance. Also, nine populations were used to assess the suitability of our most elite breeding materials for use as donors with respect to genetic background, specifically heterotic group origin, and give guidance to breeders on donor germplasm selection. Furthermore, we are studying different selection strategies for DH line breeding with respect to donor size and prior cycles of selection, i.e. recombination (none (F1), one (F2), or two selection cycles (F3)). We hope that our still ongoing research will contribute to already extensive knowledge on doubled haploid methodology.

Also, nine populations were used to assess the suitability of our most elite breeding materials for use as donors with respect to genetic background, specifically heterotic group origin, and give guidance to breeders on donor germplasm selection. Furthermore, we are studying different selection strategies for DH line breeding with respect to donor size and prior cycles of selection, i.e. recombination (none (F1), one (F2), or two selection cycles (F3)). We hope that our still ongoing research will contribute to already extensive knowledge on doubled haploid methodology.",

AB  - Овим техничким решењем описан је технолошки поступак добијања конзервисаног производа од младих клипова кукуруза baby corn обогаћеног антоцијанинима из семењаче црне соје. Добијен је стабилан функционални прехрамбени производ обогаћен биоактивним једињењима која могу позитивно утицати на здравље конзумената. Искоришћењем сојине семењаче – споредног производа из прераде соје додатно је валоризована ова сировина. Висок садржај прехрамбених влакана је такође посебно важан због неопходности повећаног уноса ових хранљивих материја у савременој исхрани због повољног утицаја на гастроинтестинално здравље. Сензорском анализом производ добијен од младих клипова хибрида кокичара са додатком 3% соли и 2% шећера такође је најбоље оцењен од стране панелиста на основу већег броја оцењиваних параметара (крцкавост, сјајност, интензитет боје, арома на ферментисано поврће). Киселост овог производа је означена као најприхватљивија (најмање кисео), што је раније потврђено и хемијском анализом pH вредности. На основу сумираних резултата, хибрид кокичар са наливом који садржи со (3%) и шећер (2%) оцењен је као оптималан за производњу пастеризованог производа од младих клипова кукуруза обогаћеног антоцијанинима екстрахованим из семењаче црне соје. Узимањем у обзир изузетно висок садржај укупних фенолних једињења, флавоноида, антоцијана, као и висок антиоксидативни капацитет, овај производ је одабран као оптималан за производњу на индустријском нивоу.
