Evaluation of genetic diversity in soybean genotypes with RAPD markers
Procena genetičke divergentnosti genotipova soje pomoću RAPD markera
Abstract
The choice of parental mating pairs is of a crucial importance in the breeding process. Obtaining a superior progeny can be expected if the parental lines, used for hybridization, are genetically different and possess as much as possible independent loci for a desirable trait, with alleles different in each parent. The utilization of genetic markers can provide the previous information on the genetic similarity of parents, and based on it, the performance of traits in the progeny can be predicted. The objective of this study was to estimate a genetic distance between the genotype ZPS 015 and genotypes L 91-3103, Ravnica, Shine, L 90-14 and ZPS 107. The genotype ZPS 015 was used as a parent in crosses toother five genotypes. All genotypes have good agronomic traits, so it was supposed that crosses among them could result in a superior progeny. Genomic DNA of these six genotypes was amplified with 18 RAPD primers. The results obtained (presence or absence of the DNA band) were transforme...d into a binary system, and were used to calculate Jaccard's similarity coefficient and a simple matching coefficient. The level of polymorphism detected (20%) and genetic distances calculated suggest a small genetic distance between parental pairs, which is in accordance with previous analyses of soybean genome polymorphism. The shortest, i.e. greatest genetic distance, by both coefficients, was determined between genotypes ZPS 015 and L 90-14, i.e. ZPS 015 and ZPS 107, respectively. The data on grain yield of the F4 progeny by each cross, obtained in the two-year trial, showed a negative and non significant correlation with the genetic distance of parents.
Izbor roditeljskih parova jedan je od ključnih momenata u procesu oplemenjivanja. Dobijanje superiornog potomstva može se očekivati ukoliko se za ukrštanje koriste genetički udaljeni roditelji koji sadrže što veći broj nezavisnih lokusa za svojstvo na koje se vrši oplemenjivanje, sa alelima različitim kod oba roditelja. Primenom genetičkih markera moguće je doći do prethodne informacije o genetičkoj sličnosti roditelja i na osnovu nje pretpostaviti osobine potomstva. U ovom radu cilj je bio ispitati genetičku distancu između genotipa ZPS 015 i genotipova L 91-3103, Ravnica, Shine, L 90-14 i ZPS 107. Genotip ZPS 015 korišćen je kao roditelj u ukrštanjima sa navedenih pet genotipova. Svi genotipovi se odlikuju dobrim agronomskim svojstvima, te je pretpostavka da će se njihovim ukrštanjem dobiti superiorno potomstvo.
