Genotypic variability of root and shoot traits of bread wheat (Triticum aestivum L.) at seedling stage

AB  - The evaluation of the embryonic root and stem of bread wheat (Triticum aestivum L.) in the early stage of development (seedling stage) can be a powerful tool in wheat breeding aimed at obtaining progenies with a greater early vigour. It is revealed that genotypes with faster early vigour have produced higher biomass and grain yield. In this study, the evaluation of traits of the embryonic root and the embryonic stem of 101 bread wheat genotypes was preformed at the 10-day old seedlings. The following eight morphological traits of roots and stems were analysed: primary root length, branching interval, the number of roots, total length of lateral roots, angle of seminal roots, stem length, root dry weight and the stem dry weight. Analysed lateral roots included seminal roots. The greatest, i.e. the smallest variability of observed traits was detected in the branching interval, i.e. the stem length, respectively. The highest positive correlation was determined between the primary root length and the total length of lateral roots. The cluster analysis, based on observed traits, shows that genotypes were clearly divided into two main clusters, A and B. The two clusters essentially differed from each other in the values of the following traits: primary root length, total length of lateral roots, root dry weight, stem dry weight and the stem length. Genotypes with shorter primary and lateral roots, lower root and stem dry weight and a shorter stem were grouped in the cluster B. On the other hand, the cluster A encompassed genotypes with values of these traits above or around the average. The values of the remaining analysed traits: the angle of seminal roots, the number of lateral roots and the branching interval varied greatly between obtained clusters. The cluster analysis showed the homogeneity of genotypes originating from Serbia and the region; their values of the root and stem length and weight were mostly around and below the average. However, the values of the angle of seminal roots, number of lateral roots and the branching interval were above average.
AB  - Ocena svojstava korena i stabla hlebne pšenice (Triticum aestivum L.) u ranoj fazi razvića, na stupnju klijanaca, može biti moćan alat u oplemenjivanju pšenice sa ciljem dobijanja potomstva sa bržim ranim porastom. Pokazano je da su genotipovi sa bržim ranim porastom proizveli veću biomasu i prinos zrna. U ovoj studiji izvršena je ocena svojstava korena i stabla 101 genotipa hlebne pšenice u fazi klijanaca starosti 10 dana. Analizirano je 8 morfoloških svojstava korena i stabla: dužina primarnog korena, distanca do prve grane na primarnom korenu, broj bočnih korenova, ukupna dužina bočnih korenova, ugao seminalnih korenova, dužina stabla, suva masa korena i suva masa stabla. The morfological and molecular identification of Fusarium verticillioides causing fusariosis on wheat grain

AB  - During the 2014-2017 period, wheat samples were collected from discoloured spikes affected by Fusarium head blight (FHB) from 20 locations in Serbia. After isolation, fungi were cultivated on potato dextrose agar (PDA) at 25oC for 7 days. Based on the in situ identification on PDA, 36 isolates of the section Liseola were selected for further analyses. The pathogenicity of all isolates was confirmed on wheat leaves. The virulence of isolates was determined by measuring the lengths of spots formed at the inoculation leaf site. In order to prove the presence of the species Fusarium verticillioides, a pair of primers FV-F2/FV-R was used. This pair of primers amplifies the sequence of the gaoB gene, and it proved to be specific for the stated species. Moreover, for the same purpose, a pair of primers VER1-VER2 based on the calmodulin partial gene was used. The reference isolate RBG 1603 Q27 was used as a positive control. The pair of primers VER1-VER2 produced a band of the expected size - 578 bp in 18 isolates, while using FV-F2/FV-R, a 370bp long band confirmed the presence of F. verticillioides in 16 samples. Sixteen out of 18 isolates verified with VER1-VER2 were also identified as F. verticillioides with FV-FS/FV-R. No amplification was observed in a negative control.
AB  - Tokom perioda od 2014. do 2017. godine sakupljani su uzorci pšenice sa fuzarioznih klasova sa 20 lokacija u Srbiji. Nakon izolacije, gljive su gajene na KDA podlozi 7 dana u termostatu na temperaturi od 25oC. Na osnovu identifikacije in situ na KDA (krompir dekstrozni agar), 36 izolata sekcije Liseola odabrano je za dalje analize. Potvrđena je patogenost svih izolata na listovima pšenice. Virulentnost izolata utvrđena je merenjem dužina formirane pege na mestu inokulacije. Za dokazivanje prisustva vrste Fusarium verticillioides korišćen je par prajmera FV-F2/FV-R koji umnožava sekvence gaoB gena i koji se pokazao kao specifičan za navedenu vrstu. Takođe, sa istim ciljem korišćen je par prajmera VER1-VER2 dizajniran na osnovu dela genske sekvence za kalmodulin. Kao pozitivna kontrola korišćen je referentni izolat RBG 1603 Q27. Par prajmera VER1-VER2 obrazovao je traku očekivane veličine (578bp) kod 18 izolata, dok je pomoću para prajmera FV-F2/FV-R traka dužine 370bp potvrdila prisustvo F. verticillioides kod 16 izolata. Šesnaest od 18 izolata koji su identifikovani VER1-VER2 parom, takođe su identifikovani i FV-FS/FV-R prajmerima. U negativnoj kontroli nije bilo amplifikacije.

AB  - Seed treatments with zinc, boron, biostimulant Coveron and MIX (zinc + boron + Coveron) were applied to three lettuce and three celeriac cultivars. Seeds of three wheat cultivars were treated under laboratory conditions with Trichoderma harzianum and eight Bacillus spp. Seed germination, seedling growth, and the presence of the following pathogens were determined: Fusarium sp., Al-ternaria sp., Penicillium sp., and Mucor sp. The Coveron treatment was the most effective on lettuce seeds tested in the germination cabinet. Seed germination was higher by 4% than in the control. Alternatively, germination of seeds treated with boron in the greenhouse was higher by 12% than in the control. The Coveron treatment had the highest effect on the shoot length, which was greater by 0.7 and 2.1 cm in the germination cabinet and the greenhouse, respectively. This treatment was also the most effective on the root length. Zn, B, and MIX treatments increased celeriac seed germination by 14% in the germination cabinet. The Zn treatment was the most efficient on seeds tested in the greenhouse. The germination was higher by 15%. A significant cultivar × treatment interaction was determined in both observed species under both conditions. The maximum effect on wheat seed germination (8%) was achieved with the T. harzianum treatment in the Salazar cultivar. A significant interdependence (p ≤ 0.01 to p ≤ 0.001) was established between seed germination and the seedling growth. The interrelationship between seed germination and pathogens of all cultivars was negative.

Alteration of metabolites accumulation in maize inbreds leaf tissue under long-term water deficit

AB  - Plants reconfigure their metabolic pathways to cope with water deficit. The aim of this study was to determine the status of the physiological parameters and the content of phenolic acids in the upper most ear leaf of maize inbred lines contrasting in drought tolerance in terms of improved plant productivity e.g., increased grain yield. The experiment was conducted under irrigation and rain-fed conditions. In drought-tolerant lines, the effect of water deficit was reflected through a chlorophyll and nitrogen balance index increase followed by a flavonols index decrease. The opposite trend was noticed in drought susceptible inbreds, with the exception of the anthocyanins index. Moreover, in comparison to irrigation treatment, opposite trends in the correlations between grain yield and physiological parameters found under water deficit conditions indicated the activation of different metabolic pathways in defense against water deficit stress. Concerning phenolic acid content, water deficit caused the reduction of protocatechuic, caffeic, and sinapic acid in all inbreds evaluated. However, the highly pronounced increase of ferulic and especially cinnamic acid content under water deficit conditions indicated possible crucial role of these secondary metabolites in preventing the harmful effects of water deficit stress, which, in turn, might be useful in maize breeding selection for drought tolerance

The aim of this study was to determine the status of the physiological parameters and the content of phenolic acids in the upper most ear leaf of maize inbred lines contrasting in drought tolerance in terms of improved plant productivity e.g., increased grain yield. The experiment was conducted under irrigation and rain-fed conditions. In drought-tolerant lines, the effect of water deficit was reflected through a chlorophyll and nitrogen balance index increase followed by a flavonols index decrease. The opposite trend was noticed in drought susceptible inbreds, with the exception of the anthocyanins index. Moreover, in comparison to irrigation treatment, opposite trends in the correlations between grain yield and physiological parameters found under water deficit conditions indicated the activation of different metabolic pathways in defense against water deficit stress. Concerning phenolic acid content, water deficit caused the reduction of protocatechuic, caffeic, and sinapic acid in all inbreds evaluated. However, the highly pronounced increase of ferulic and especially cinnamic acid content under water deficit conditions indicated possible crucial role of these secondary metabolites in preventing the harmful effects of water deficit stress, which, in turn, might be useful in maize breeding selection for drought tolerance",

AB  - Due to the loss of agro-biodiversity, there is a strong effort to find apparent and efficient
mechanisms for the conservation and sustainable use of genetic diversity. A joint monitoring of the
diversity and collections structure of the Montenegrin maize landraces conserved in the Serbian
(MRIZPGB) and Montenegrin (MGB) gene banks has been conducted in order to improve the
composition of the collections and to identify and eliminate possible redundancy. Based on a separate
analysis of white- and yellow-orange maize landraces, it can be concluded that the diversity and
evolution of distinct maize landraces grown and collected in Montenegro have been simultaneously
shaped by both environmental (i.e., natural selection) and socially driven factors (farmers’ selection,
migration and colonization processes of the human population). Although it has been determined
that the authenticity and variability of the Montenegrin maize landraces gene pool have largely
been preserved in the MRIZPGB collection, a significant amount of redundancy was observed. The
obtained results will contribute to the cost-efficient conservation of the maize gene pool in the
Montenegrin and Serbian gene banks. The recognized and well-preserved original variability of the
MRIZPGB and MGB Montenegrin gene pool represents a valuable source for pre-breeding activities
on broadening the white and flint maize breeding programmes under temperate conditions.

Black soya bean and black chia seeds as a source of nutrients and bioactive compounds with health benefits

AB  - Recent trends in healthy lifestyle and diet made functional foods rich in
quality nutrients and bioactive compounds with potential health benefits highly sought
after. Some cultivated plants, such as soya and chia can provide a viable source of
nutraceuticals with high fibre, protein, and protective antioxidant potential. The wholegrain flours of black soya beans and black chia seeds were used in this study. Potential
nutritive and health-benefitting properties of these flours were compared by assessing
their chemical composition and antioxidant profile. The content of dietary fibres such
as NDF, ADF, ADL and hemicellulose determined in black chia seed sample was
higher than in black soya soya beans which had higher fibre content. The total protein
content recorded in black soya beans was almost as twice as high (42.26+0.14%) as
chia protein content (25.04+0,20%). Black soya beans had the highest content of water
soluble proteins (29.00+0.13% d.m.) with NSI (nitrogen solubility index) acounting
for as much as 70.96+0.31% of total proteins, while black chia seeds had the highest
content of globulins (14.64+0.07% d.m.) and NSI 58.48+0.27% of total proteins. Both
black soya bean and black chia contained a high amount of total phenolic compounds
(830,66+5.46 and 1201.94+16.29 mg GAE/kg, respectively) and exhibited a
considerable total antioxidant capacity, which makes them good contestants for
functional food ingredients with potential health benefits.

AB  - The objective of this study was to evaluate the impact of two foliar fertilizers applied on five
maize (Zea mays L.) lines. Fertilizers were applied at different growth stages of maize, during three consecutive years (2010–2012) at the experimental field of the Maize Research Institute “Zemun Polje”, Serbia. Maize growth parameters such as fresh matter, height, leaf area and grain yield were recorded. Foliar fertilizer with amino acids (FAA) was more advantageous to maize plants compared to fertilizer containing phosphorus (FP) as a main component. Applied FAA has shown positive effects by increasing fresh matter, leaf area index, and plant height in all three years. In 2012, due to unfavorable meteorological conditions, grain yield and harvest index were very low, compared to the previous two years, although, positive effects on morphological traits were observed 21 days after treatments (DAT), as well as in the anthesis stage. The best results of 30% of grain yield and harvest index increase were recorded in line L1 in 2010 and 2011. The same line had an increase of more than 40% of fresh matter and leaf area on average for all three years. The positive effects that have been noticed in this research could recommend foliar fertilizing with fertilizer containing N in a form of an amino acids complex.

AB  - A soil tillage system adjusted to the soil type and agro-ecological conditions, togetherwith the optimal nutrient supply, is a prerequisite for high maize (Zea mays L.) yield. However,there is little knowledge about the influence of soil tillage and nitrogen (N) rates on maize grainquality. A study was initiated in 1978 in Zemun Polje, Belgrade, Serbia, on the chernozem soil type.The effects of three N rates—0, 180 and 240 kg N ha−1—within three tillage practices—no-tillage,reduced and conventional tillage—and their influence on grain yield (GY) and the content of proteins,starch, oil, total tocopherols and carotenoids, phytic phosphorus (Phy), glutathione (GSH), phenolsand inorganic P (Pi) in maize grain were analysed in the period 2016–2018. Differences in maizeGY were 2.57 and 3.01 t ha−1for reduced- or no-tillage, respectively, in comparison to conventionaltillage. Conventional tillage and higher N rates contributed to the increase in maize yield and thegrain nutritional quality with regard to the increased level of proteins, GSH and Phy. On the otherhand reduced inputs were beneficial only for grain quality, in relation to the increase in tocopherolsand phenols.

AB  - Savremeni trendovi u oplemenjivanju kukuruza idu u pravcu identifikacije genotipova povećanog kvaliteta zrna za ljudsku upotrebu, industrijsku preradu i njihovog uključivanja u komercijalne oplemenjivačke programe. Za ovo istraživanje odabrane su tri hibridne i tri slobodnooprašujuće sorte kukuruza različite po boji i tipu zrna, u cilju ispitivanja razlika u sadržaju ukupnih karotenoida, fenolnih jedinjenja i ukupnom antioksidativnom kapacitetu. Dodatno su analizirane fizičke karakteristike i osnovni hemijski sastav zrna, kao i sadržaj proteinskih frakcija. Multivarijacioni pristup, kroz primenu analize glavnih komponenata (PCA) doprineo je boljem razumevanju složenih veza ispitivanih parametara kvaliteta. Sa aspekta različite upotrebne vrednosti, bolji tehnološki kvalitet poželjan za proces suvog mlevenja i proces proizvodnje grickalica (snek proizvoda) je identifikovan kod slobodnooprašujućih sorti Osmak i Bosanac, dok se sorta Rumenka odlikovala visokim sadržajem bioaktivnih jedinjenja (tj. visokim antioksidativnim kapacitetom). Dobijeni rezultati ukazuju na superiornost slobodnooprašujućih sorti u poređenju sa hibridnim.

Za ovo istraživanje odabrane su tri hibridne i tri slobodnooprašujuće sorte kukuruza različite po boji i tipu zrna, u cilju ispitivanja razlika u sadržaju ukupnih karotenoida, fenolnih jedinjenja i ukupnom antioksidativnom kapacitetu. Dodatno su analizirane fizičke karakteristike i osnovni hemijski sastav zrna, kao i sadržaj proteinskih frakcija. Multivarijacioni pristup, kroz primenu analize glavnih komponenata (PCA) doprineo je boljem razumevanju složenih veza ispitivanih parametara kvaliteta. Sa aspekta različite upotrebne vrednosti, bolji tehnološki kvalitet poželjan za proces suvog mlevenja i proces proizvodnje grickalica (snek proizvoda) je identifikovan kod slobodnooprašujućih sorti Osmak i Bosanac, dok se sorta Rumenka odlikovala visokim sadržajem bioaktivnih jedinjenja (tj. visokim antioksidativnim kapacitetom). Dobijeni rezultati ukazuju na superiornost slobodnooprašujućih sorti u poređenju sa hibridnim.",
