Seed quality of reciprocal maize single crosses and their parental inbred lines

AB  - Five direct and reciprocal early hybrids, five direct and reciprocal late hybrids and a total
of their 12 parental inbred lines were used in order to test reciprocal effects among
hybrids in early germination phase and seed quality of their parental inbred lines. Five
hybrids were of early maturity FAO 300-400 group, and five of late maturity FAO 500-
600 group. In the early group one line of Lancaster origin (L1) was in common, and in
direct crosses used as male parent, while in reciprocal crosses as female parent. The same
case was with late hybrids where another Lancaster line (L2) was in common. In 2015
and 2016 all the material was multiplied in hand pollination and after six months of
storage three tests according to ISTA standards were performed: standard germination test
(SGT), cold-test (CT) and vigor radicle test (RE). According to the Analysis of variance,
a strong reciprocal effect was found among early hybrids for four out of five parameters
in RE and SGT, while it was found only for RE in late hybrids. However, in CT none of
the parameters had significant reciprocal effect. Two tested Lancaster inbreds were
superior in the hybrids over non-Lancaster lines when used as maternal components,
while as lines per se they did not display such superiority. Mid-parent heterosis (MPH)
ranged from -62.32% to 211.11% in RE test, with an average of 44.26%; from -10.15% to
17.93% in SGT test, with an average of 6.27%, and from -5.31% to 4.49% in CT, with an
average of 0.64%. Significant differences of MPH were found for nine pairs of reciprocal
hybrids for RE, three pairs for SGT and seven pairs for CT. This shows that reciprocal
differences in MPH were higher as the applied level of stress during early seed growth
raised. All the results showed very complicated mode of inheritance of tested traits in the
early developmental stages of young plants, including the phenomenon of heterosis which
is expressed already at the germination stage.

AB  - In vivo metoda dupliciranih haploida u Institutu za kukuruz „Zemun Polje“ koristi se u oplemenjivanju kukuruza od 2014. godine. Pristup omogućava dobijanje potpuno homozigotnih inbred linija tzv. duplohaploidnih (DH) linija za samo dve godine (dve do tri generacije), za razliku od konvencionalnog oplemenjivanja gde je potrebno bar sedam do osam generacija kako bi se postigla neophodna homozigotnost. Metoda se sastoji iz nekoliko koraka: 1) indukcija haploida iz odabranih populacija ukrštanjem sa linijom induktorom haploida (prva godina/letnja generacija); 2) selekcija haploidnih zrna (prva godina/letnja generacija); 3) duplikacija hromozoma (druga godina/zimska generacija) i 4) umnožavanje dobijenih DH linija (druga godina/letnja generacija). Do sada je dobijeno oko 13000 DH linija iz oko 350 populacija koje pripadaju različitim heterotičnim grupama. U okviru programa sprovode se i aktivnosti sa ciljem razvoja novih induktora haploida sa povišenim procentom indukcije haploida i poboljšanim agronomskim svojstvima. Osam induktora-kandidata je specifičnom procesom selekcije odabrano i u 2023. godini ocenjeno u mini ogledu sa komercijalno dostupnim induktorima, a mereni su visina biljke, produkcija polena, ozrnjenost u ukrštanju, kao i procenat indukcije haploida. U okviru rada na optimizaciji metode, ispitivan je i uticaj populacije (heterotične grupe) i spoljašnje sredine na uspešnost metode. Utvrđen je značajan uticaj spoljašnjih uslova, kao i heterotične grupe odnosno genotipa na indukciju i duplikaciju, kao i konačnu uspešnost metode. Prvi hibridi sa DH linijom kao roditeljskom komponentom već su registrovani od strane Sortne komisije Republike Srbije za komercijalno gajenje u Srbiji.

AB  - U Institutu za kukuruz "Zemun Polje" postoji program selekcije pomoću molekularnih markera (MAS) čiji je cili ugradnja gena za inkompatibilnost Ga1-S u linije kukuruza koje su roditeljske komponente hibrida belog zrna. Glavni cilj ovog rada je bio stvaranje linija kukuruza belog zrna sa potpunom cross-inkompatibilnošću sa stranim polenom pomoću molekularnih makera. Molekularnim markerima koji su blisko vezani za Ga1-S gen uspešno su identifikovane heterozigotne bilike (50%) nakon sva tri povratna ukrštanja. Rezultati utvrđivanja procenta genoma rekurentnog roditelia (RPG) u BC3 generaciji pokazali su vrednost iznad očekivane kod 69% potomstva, dok je kod određenog broja RPG iznosio 99% što predstavlja vrednost koja se teoretski dostiže u BC6 generaciji. Od 264 pojedinačne bilike BC3F2 generacije, 70 (26,5%) su bili dominantni homozigoti, što je u skladu sa očekivanim prema pravilima Mendelovog nasleđivanja. Potomstva BC3F2, dominantno homozigotnih biljaka posejana su sa kukuruzom žutog zrna iste grupe zrenja radi provere cross-inkompatibilnosti. Nijedno od dobijenih potomstava nije imalo 100% belo zrno, najverovatnije usled prisustva gena modifikatora koji menjaju efikasnost isključivanja polena, ili je došlo do gublienja ovih gena prilikom povratnog ukrštanja. Moglo bi se očekivati da bi se primenom funkcionalnih markera (stvorenih na osnovu polimorfizama sekvenci unutar samog gena vezanog za fenotipsku osobinu) uspešno prevazišle uočene prepreke i poboljšala efikasnost MAS za prenos željenih gena koji kontrolišu prostu ili složenu osobinu u gajene kulture.

Adapted Quality Protein Maize in Broiler Diets Holds Nutritive and Financial Advantages

AB  - Quality protein maize (QPM) can be beneficial for use in feeds as it was shown that substitution of standard maize (SM) with QPM can improve livestock performance and decrease costly dietary lysine supplementation and protein ingredients. Herein, nutritional and financial effects of QPM adapted to temperate climate in broiler diets were tested. Feeding experiment was performed on two groups of broilers – control (fed with SM) and treatment (fed with QPM). Duration of each feeding trial was 42 days, comprising three phases - starter (1-14), grower (15-35) and finisher (36-42). Diets for all growth phases were formulated based on the biochemical analysis of SM and QPM kernels. Body weight of the chickens was measured on days 1, 7, 14, 35 and 42. Feed intake (FI), body weight gain (BWG), average daily gain (ADG) and feed conversion ratio (FCR) were calculated at the end of each phase. Two trials were performed. In the first, SM was completely replaced with QPM in treatment group diets. As the results indicated better FCR in the treatment group for grower (1.49:1.54) and finisher (1.55:1.60) phases, in the second trial QPM was increased and soybean component decreased by 3%. That led to better results as FCR was even lower but in all three phases, confirming the results of the first trial and further confirming the effects of QPM on feed conversion. Moreover, significant financial reduction in feed was observed, as the price of soybean is usually two to four times higher than the price of maize.

AB  - The aim of this study was to assess the chemical composition and bioactive properties of the wholegrain flour obtained from eleven maize inbred lines to identify genotypes with increased potential for the development of hybrids with high nutritional and functional value, suitable for food production. The maize inbreds, including seven standard yellow, two QPM (quality protein maize) and two lines for red kernel hybrids, were grown in the experimental field of the Maize Research Institute at the location of Zemun Polje, Serbia. Wholegrain maize flour was got by grinding the maize grain in a laboratory mill. The assessment of the chemical composition and content of certain bioactive compounds, as well as the total antioxidant capacity, was conducted using standard laboratory procedures. The highest starch content (73.73%) was determined in line L8, while line L10 had the highest protein content (12.82%). Among soluble proteins, the α-zein fraction was dominant in most of the lines, ranging from 0.92% to 3.57%. The highest content of total fibres (NDF) was determined in red kernel line L9 (15.77%). Line L8 was the richest in total carotenoids (21.08 μg βCE/g d.m.), while line L7 had the highest total antioxidant capacity (34.30 mmol Trolox/kg d.m.), which can be explained by the presence of anthocyanins in the red grain. Line L1 had the highest content of total sugars (3.36%), and line L4 had the lowest (1.44%). Most of the samples of inbred lines investigated in this study showed good quality parameters regarding chemical composition and bioactive properties. The obtained results may provide some valuable guidelines needed in the following stages of maize breeding and open up various possibilities for the utilization of wholegrain maize flour in the food industry.
AB  - Cilj ovog istraživanja bio je da se odrede hemijski sastav i bioaktivna svojstva integralnog brašna dobijenog od jedanaest inbred linija kukuruza u cilju identifikacije genotipova sa povećanim potencijalom za razvoj hibrida visoke nutritivne i funkcionalne vrednosti, pogodnih za proizvodnju hrane. Linije kukuruza, uključujući sedam standardnih žutih, dve QPM i dve linije za hibrid crvenog zrna, gajene su na oglednom polju Instituta za kukuruz na lokaciji Zemun Polje, Srbija. Integralno kukuruzno brašno dobijeno je mlevenjem u laboratorijskom mlinu. Procena hemijskog sastava i sadržaja pojedinih bioaktivnih jedinjenja, kao i ukupnog antioksidativnog kapaciteta, sprovedena je primenom standardnih laboratorijskih procedura. Najviši sadržaj skroba (73,73%) utvrđen je u liniji L8, dok je linija L10 imala najveći sadržaj proteina (12,82%). Među rastvorljivim proteinima, frakcija α-zeina bila je dominantna u većini linija, u rasponu od 0,92% do 3,57%. Najveći sadržaj ukupnih vlakana (NDF) utvrđen je u zrnu crvene boje Linije 9 (15,77%). Linija L8 je bila najbogatija ukupnim karotenoidima (21,08 mg βCE/g s.m.), dok je linija L7 imala najveći ukupni antioksidativni kapacitet (34,30 mmol Trolox/kg d.m.), što se može objasniti prisustvom antocijana u crvenom zrnu. Linija L1 je imala najveći sadržaj ukupnih šećera (3,36%), a linija L4 imala je najmanji (1,44%). Svi uzorci novih inbred linija ispitivani u ovoj studiji pokazali su dobre parametre kvaliteta u pogledu hemijskog sastava i bioaktivnih svojstava. Dobijeni rezultati mogu dati neke dragocene smernice potrebne u narednim fazama oplemenjivanja kukuruza, kao i otvoriti različite mogućnosti za korišćenje integralnog kukuruznog brašna u prehrambenoj industriji.

Svi uzorci novih inbred linija ispitivani u ovoj studiji pokazali su dobre parametre kvaliteta u pogledu hemijskog sastava i bioaktivnih svojstava. Dobijeni rezultati mogu dati neke dragocene smernice potrebne u narednim fazama oplemenjivanja kukuruza, kao i otvoriti različite mogućnosti za korišćenje integralnog kukuruznog brašna u prehrambenoj industriji.",

AB  - The objective of this study was to early identify genotypes with increased potential for the development of hybrids with high nutritional and functional value, suitable for food production, by assessing the grain quality parameters of eleven new maize inbred lines. The inbreds, including nine standard yellow and two red kernel lines, were grown in the experimental field of the Maize Research Institute at the location of Zemun Polje, Serbia in 2020. Wholegrain maize flour was obtained by a dry grind process on a laboratory mill. The assessment of the chemical composition and content of certain bioactive compounds, as well as total antioxidant capacity, was conducted using standard laboratory procedures. The highest starch content (73.73%), was determined in Line 8, while Line 10 had the highest protein content (12.82%). The lowest oil content was determined in red kernel Line 7, namely 4.87%. Among soluble proteins, the α-zein fraction was dominant in most of the lines, ranging from 0.92% to 3.57%. The glutelin and albumin fraction were present in a lower percentage, followed by the globulin fraction. The highest content of total fibers (NDF) was determined in red kernel Line 9 (15.77%). Line 8 was the richest in total carotenoids (21.08 μg βCE/g d.m.), while Line 7 had the highest total antioxidant capacity (34.30 mmol Trolox/kg d.m.), which can be explained by the presence of anthocyanins in the red grain. Line 1 had the highest content of total sugars (3.36%), and Line 4 had the lowest (1.44%). All samples of new inbred lines investigated in this study showed good quality parameters regarding chemical composition and bioactive properties. The obtained results may provide some valuable guidelines needed in the following stages of maize breeding as well as open up various possibilities for the utilization of wholegrain maize flour in the food industry.

AB  - Red and blue colors in maize kernel are the consequence of anthocyanin pigments, responsible for the increased phenolic content and antioxidant activity in maize. Therefore, interest in pigmented maize as a novel functional food due to its possible health benefits and unique taste has grown. The red color in maize kernel originates from pericarp and is maternally inherited while the blue color originates from aleurone, which is triploid in nature and its mode of inheritance is more complicated. Red and blue maize are usually low-yielding populations, and at the Maize Research Institute Zemun Polje (MRIZP), commercial inbred lines have been converted to red and blue colors to produce high-yielding red and blue maize hybrids.
Maize extractable phenolic content (EPC) can be directly determined through wet chemistry and measures phenolic amides, glycerides, free phenolic acids, and some flavonoids. In this study, however, Fourier-transformed near-infrared reflectance spectroscopy (FT-NIRS), as a non-destructive, inexpensive, and rapid technique has been used to scan EPC profiles of 56 MRIZP maize samples (14 genotypes in two replications and two environments) differing in kernel color: yellow, red pericarp, blue aleurone and red pericarp with blue aleurone. Twenty samples with outlying EPC values have been directly analyzed for total phenolics and these values have been used to adjust and improve NIRS calibration.
EPC values ranged from 1.69 to 6.82 mg g-1 across genotypes and environments. Two red inbred lines had the highest EPC content in 2017, 6.82 and 5.11 mg g-1, and, overall samples from 2017 had a higher content of EPC compared to the same genotypes grown in 2018. Red genotypes on average had the highest EPC content, followed by red-blue, while yellow and blue kernels had similar and the lowest content within the samples.

AB  - Limited water availability and nitrogen deficiency are the most restricting factors for maize growth. Although maize susceptibility to drought is the highest at the reproductive stage, water shortage during vegetative development can significantly reduce grain yield. The aim of this study was to assess the genetic variation of growth dynamics in 20 maize inbred lines (ILs) through automated phenotyping based on visible light (RGB) imaging in a greenhouse under optimal (C), nitrogen (N), water (W), and combined nitrogen and water stress (NW) conditions. Thirteen biomass-related and morpho-physiological image-derived traits acquired at 33 time points were selected, covering developmental stages from five fully developed leaves to 10-13 fully developed leaves, depending on treatment. Estimated biomass volume (EBv), a proxy for biomass in our study, showed to be the most sensitive image-derived morphological trait to water and combined water and nitrogen stress with the maximum reduction of 53 and 54% in W and NW compared to C at the time of maximum water stress. Genotypic variation of EBv within each treatment was consistently high and over 80%, while the analysis across treatments showed that as stress got higher the genetic variation got lower (32-38%) and the interaction term became more prominent. Phenotypic correlations between EBv and other image-derived traits at early stages showed that under no stress or mild stress conditions, morphological traits were more appropriate than color-related traits for the prediction of biomass accumulation, while under more severe stress conditions, color-related traits and chlorophyll fluorescence are more useful to differentiate genotypes for high biomass. To investigate whether ILs can express EBv in similar patterns, temporal profiles were clustered by using the fuzzy c-means clustering algorithm, and two temporal dynamics of EBv patterns among the studied ILs in each treatment were identified.

AB  - Small non-coding RNAs (sRNAs) have an important role in regulating plant gene expression,
both in developmental processes and stress responses, through RNA interference or silencing.
sRNAs respond to environmental stressors in manners dependant on the type of stress, tissue
and genetic background. Herein, their role in chilling stress response is examined in young maize
seedlings (Zea mays L.) of two inbred lines of contrasting chilling tolerance, using high-throughput
sequencing methods. Understanding this response is critical since one of the most promising
strategies of avoiding maize yield loss due to climate change is earlier sowing, during periods
of suboptimal temperatures. Sterilized seeds of the two lines, marked as LT (tolerant), and LS
(sensitive), were germinated in the dark for five days (optimal conditions), after which the 5-d
old seedlings were exposed to chilling conditions for 24h (temperature: 10/8 °C; photoperiod:
12/12h). Samples for RNA isolation were taken after 6h and 24h of treatment. After the cDNA

Significant number of those circRNA have shown expression dependent on chilling, as well as genotype, but further studies are necessary to confirm their role in this process, find their target molecules and unravel regulatory networks they create with other types of RNAs.

AB  - Reciprocal effect in maize refers to the phenotypic difference between reciprocal F1 hybrids.
The aim of this study was to estimate influence of reciprocal crosses on grain yield and eight
yield components. Five single-cross hybrids, their reciprocal crosses and six parental inbred
lines were selected and all hybrids belong to late maturity group (FAO 500-600) with Lancaster
ZPL-7 line as mutual parent. The experiment was set up in two replications using a completelyrandomized block design in 2016 and 2017, at a total of seven environments. Reciprocal effect
significantly influenced only grain yield and ear length, while factors hybrid and location were
significant for all the examined traits. All hybrids individually displayed significant reciprocal
effect for grain yield, ear length, number of kernels per row and 100 kernel weight, two hybrids
for kernel length and thickness, while three hybrids showed reciprocal effect on kernel width.
The effects were both positive and negative depending on the genotype itself. Furthermore,
there were no significant differences for ear row number and ear width between normal and
reciprocal variants. The highest difference for grain yield was between ZP 606 reciprocal
(12.06 t/ha) and ZP 606 normal (11.28 t/ha). Pearson correlations were calculated between
examined traits. All correlations between grain yield and other yield components were found
significant and positive, except for the number of rows per ear (no correlation) and kernel width
(negative correlation). Reciprocal effect has strong influence on the measured yield traits, but
it is genotype specific. Therefore, in the future we should examine normal and reciprocal
variants of all commercial maize hybrids.

AB  - ZPQPM13 is a maize hybrid with improved protein quality, adapted to temperate climate growth conditions, developed by crossing a QPM version of ZPL5 commercial line (developed by marker assisted selection) and GS-6 (a QPM line with 50% tropical and 50% temperate germplasm). This hybrid has grain yield as commercial hybrids, over 90% of hard endosperm, contents of tryptophan and protein over 0.075% and 10%, respectively, and stability of tryptophan content over different locations and years. ZPQPM13 was tested for use in broiler feeds, with the aim to reduce utilization of costly synthetic lysine or soybean components. The feeding experiment on Ross 308 broilers was divided into two groups – control (fed with standard maize) and treatment (fed with QPM). In treatment group diets, ZPQPM13 was increased and soybean component decreased for 3%. Chemical analyses revealed higher contents of limiting essential amino acids in mercantile ZPQPM13 compared to standard maize – 0.44 vs. 0.25 % lysine, 0.077 vs. 0.070 % tryptophan and 0.39 vs. 0.27 % threonine. Duration of each feeding trial was 42 days, comprising three phases - starter (1-14 days), grower (15-35 days) and finisher (36-42 days). Feed intake (FI), body mass gain (BMG), average daily gain (ADG) and feed conversion ratio (FCR) were calculated at the end of each phase. In treatment group, FI was reduced, while BMG and ADG were higher in starter phase but lower in grower and finisher phases. FCR was lower in all three phases in treatment group – 1.13 vs. 1.30, 1.25 vs. 1.33 and 1.28 vs. 1.45. Although a small drawback is that final weight was 3.4 % higher in control broilers, comparison of feed and meat prices show that financial benefits of using QPM are still significant compared to using standard maize in feeds, as soybean is four times more expensive than maize.

Surveying maize seedlings as the first step for re-evaluating cold tolerance in maize inbred lines

AB  - The consequences of the current climate changes sustain the strategy of early sowing (early spring) to avoid drought during the flowering
period and thus secure maize yields. Earlier sowing dates increase the risk of exposure of the plants to cold during the early development stages
and thus re-evaluation of elite material is required to select more tolerant genotypes. The first step of re-evaluation is to define characteristics that
could be used for fast survey of the elite material, before performing more profound analyses. The aim of this paper was to analyse morphological
characteristics (radicle length, coleoptile length, seedling weight) of 5 day old seedlings of two inbred lines LK and Mo17 (according to breeders
experience tolerant and susceptible, respectively) subjected to chilling stress (10°C day/8°C night) for 24h and 48h, as well as to perform
morphological (root and shoot characteristics) and physiological (chlorophyl, flavonoid and anthocyanin contents) analyses of the plants after
seven day recovery. Control plants were grown under optimal conditions (30°C day /20°C night, ≈60% humidity), at an 8h photoperiod. The
results were not straightforward, as the susceptible Mo17 showed faster recovery (statistically insignificant differences between control and
treatment plants for most of the analysed traits), while the tolerant LK was a more robust genotype throughout the experiment. Chlorophyl,
flavonoid and anthocyanin contents were not informative. The results identified survival rate and root characteristics as informative for fast
screening of seedlings for determination of cold tolerance.
