MARKER-ASSISTED GА1-S INCORPORATION INTO WHITE MAIZE HYBRID´S PARENTAL LINES

AB  - Maize Research Institute Zemun Polje has a marker assisted backcross breeding (MABB) program aimed to incorporate the strong allele of incompatibility gene Gа1 into parental components of the hybrids with specific traits (white kernel). The main objective of this research was marker assisted development of the white maize hybrid´s parental lines with the complete cross-incompatibility to foreign pollens. Ga1-S-tightly-linked molecular markers successfully identified heterozygous plants with the percentage of approximately 50% after all three backcrossing. Regarding the background selection, 69% of progenies had recurrent parent’s genome (RPG) above the theoretical value for BC3 generation. Also, a few individuals even had 99%, the value theoretically achieved in BC6 generation, showing that MABB made a genetic gain in RPG recovery. Foreground selection in this generation aimed to identify homozygous dominant individuals. Out of 264 BC3F2 plants, 70 (26.5%) were dominant homozygotes, which is in accordance with the expected Mendelian ratio. Progenies of the BC3F2 homozygous dominant plants were planted alternatively with yellow-kernel maize of the same maturity to check cross-incompatibility. Unfortunately, none of the dominant progenies had a 100% white kernel, most likely due to the presence of modifier genes that increase/decrease the effectiveness of pollen exclusion or that, alternatively, modifiers were lost during the backcrossing. It could be expected that successfully employed functional markers (developed from the sequence polymorphisms present within functional gene(s) associated with phenotypic trait variations) would outbalance the noted impediments and enhance MABB efficiency to transfer the desired gene(s) controlling simple or complex trait(s) into cultivated varieties.
AB  - U Institutu za kukuruz “Zemun Polje” postoji program selekcije pomoću molekularnih markera (MAS) čiji je cilj ugradnja gena za inkompatibilnost Gа1-S u linije kukuruza koje su roditeljske komponente hibrida belog zrna. Glavni cilj ovog rada je bio stvaranje linija kukuruza belog
zrna sa potpunom unakrsnom inkompatibilnošću sa stranim polenom pomoću molekularnih
makera. Molekularnim markerima koji su blisko vezani za Ga1-S gen uspešno su identifikovane
heterozigotne biljke (50%) nakon sva tri povratna ukrštanja. Rezultati utvrđivanja procenta genoma rekurentnog roditelja (RPG) u BC3 generaciji pokazali su vrednost iznad očekivane kod 69%

zrenja radi provere unakrsne inkompatibilnosti. Nijedno od dobijenih potomstava nije imalo 100%
belo zrno, najverovatnije usled prisustva gena modifikatora koji menjaju efikasnost isključivanja
polena, ili je došlo do gubljenja ovih gena prilikom povratnog ukrštanja. Moglo bi se očekivati da
će uspešna primena funkcionalnih markera (stvorenih na osnovu polimorfizama sekvenci unutar
samog gena vezanog za fenotipsku osobinu) prevazići uočene prepreke i poboljšati efikasnost MAS
za prenos željenih gena koji kontrolišu prostu ili složenu osobinu u gajene kulture.

Tryptophan content increase in maize inbred lines improved through marker assisted breeding

AB  - Quality protein maize (QPM) has high content of essential amino acids lysine and tryptophan that are deficient in standard maize. Naturally occurring opaque2 recessive mutation has been the most widely studied and used as a source for genetic improvement of the nutritional value of maize proteins. Marker assisted conversion of maize inbred lines to QPM adapted to temperate regions is being conducted at Maize Research Institute Zemun Polje (MRIZP). The ultimate goal is selection of the opaque2 recessive genotypes (foreground selection) with the highest proportion of recurrent parentʹs genome (background selection) along with high lysine/tryptophan content without losing good agronomic performances of the original line. After two backcrossing and selection of heterozygous individual plants, opaque2-specific molecular markers phi057 and umc1066 successfully identified recessive homozygotes in BC2F2 generation. The next step was to confirm expression of the QPM trait through elevated kernel tryptophan content (TC). Laboratory analyses were used to quantify and select for acceptable tryptophan concentrations for QPM - above 0.075%. The results presented in this paper relate to identification of BC2F3 progenies with increased TC in the conversion of two MRIZP commercial inbred lines (RP1 and RP2). The average TC ranged from 0.070% in RP1 to 0.074% in RP2. Out of 60 progenies from these two lines, 27 had TC at or above the QPM treshold. A total of 12 progenies was chosen for the highest tryptophan content. These lines will serve as parental lines for developing QPM hybrids adapted to temperate regions.

Progenies of the BC3F2 homozygous dominant plants were planted alternatively with yellow-kernel maize of the same maturity to check cross-incompatibility. Unfortunately, none of the dominant progenies had a 100% white kernel, most likely due to the presence of modifier genes that affect the effectiveness of pollen exclusion or that, alternatively, modifiers were lost during the backcrossing. It could be expected that successfully employed functional markers (developed from the sequence polymorphisms present within functional gene(s) associated with phenotypic trait variations) would outbalance the noted impediments and enhance MABB efficiency to transfer the desired gene(s) controlling simple or complex trait(s) into cultivated varieties.",

AB  - U Institutu za kukuruz "Zemun Polje" postoji program selekcije pomoću molekularnih markera (MAS) čiji je cili ugradnja gena za inkompatibilnost Ga1-S u linije kukuruza koje su roditeljske komponente hibrida belog zrna. Glavni cilj ovog rada je bio stvaranje linija kukuruza belog zrna sa potpunom cross-inkompatibilnošću sa stranim polenom pomoću molekularnih makera. Molekularnim markerima koji su blisko vezani za Ga1-S gen uspešno su identifikovane heterozigotne bilike (50%) nakon sva tri povratna ukrštanja. Rezultati utvrđivanja procenta genoma rekurentnog roditelia (RPG) u BC3 generaciji pokazali su vrednost iznad očekivane kod 69% potomstva, dok je kod određenog broja RPG iznosio 99% što predstavlja vrednost koja se teoretski dostiže u BC6 generaciji. Od 264 pojedinačne bilike BC3F2 generacije, 70 (26,5%) su bili dominantni homozigoti, što je u skladu sa očekivanim prema pravilima Mendelovog nasleđivanja. Potomstva BC3F2, dominantno homozigotnih biljaka posejana su sa kukuruzom žutog zrna iste grupe zrenja radi provere cross-inkompatibilnosti. Nijedno od dobijenih potomstava nije imalo 100% belo zrno, najverovatnije usled prisustva gena modifikatora koji menjaju efikasnost isključivanja polena, ili je došlo do gublienja ovih gena prilikom povratnog ukrštanja. Moglo bi se očekivati da bi se primenom funkcionalnih markera (stvorenih na osnovu polimorfizama sekvenci unutar samog gena vezanog za fenotipsku osobinu) uspešno prevazišle uočene prepreke i poboljšala efikasnost MAS za prenos željenih gena koji kontrolišu prostu ili složenu osobinu u gajene kulture.

Adapted Quality Protein Maize in Broiler Diets Holds Nutritive and Financial Advantages

AB  - Quality protein maize (QPM) can be beneficial for use in feeds as it was shown that substitution of standard maize (SM) with QPM can improve livestock performance and decrease costly dietary lysine supplementation and protein ingredients. Herein, nutritional and financial effects of QPM adapted to temperate climate in broiler diets were tested. Feeding experiment was performed on two groups of broilers – control (fed with SM) and treatment (fed with QPM). Duration of each feeding trial was 42 days, comprising three phases - starter (1-14), grower (15-35) and finisher (36-42). Diets for all growth phases were formulated based on the biochemical analysis of SM and QPM kernels. Body weight of the chickens was measured on days 1, 7, 14, 35 and 42. Feed intake (FI), body weight gain (BWG), average daily gain (ADG) and feed conversion ratio (FCR) were calculated at the end of each phase. Two trials were performed. In the first, SM was completely replaced with QPM in treatment group diets. As the results indicated better FCR in the treatment group for grower (1.49:1.54) and finisher (1.55:1.60) phases, in the second trial QPM was increased and soybean component decreased by 3%. That led to better results as FCR was even lower but in all three phases, confirming the results of the first trial and further confirming the effects of QPM on feed conversion. Moreover, significant financial reduction in feed was observed, as the price of soybean is usually two to four times higher than the price of maize.

AB  - Limited water availability and nitrogen deficiency are the most restricting factors for maize growth. Although maize susceptibility to drought is the highest at the reproductive stage, water shortage during vegetative development can significantly reduce grain yield. The aim of this study was to assess the genetic variation of growth dynamics in 20 maize inbred lines (ILs) through automated phenotyping based on visible light (RGB) imaging in a greenhouse under optimal (C), nitrogen (N), water (W), and combined nitrogen and water stress (NW) conditions. Thirteen biomass-related and morpho-physiological image-derived traits acquired at 33 time points were selected, covering developmental stages from five fully developed leaves to 10-13 fully developed leaves, depending on treatment. Estimated biomass volume (EBv), a proxy for biomass in our study, showed to be the most sensitive image-derived morphological trait to water and combined water and nitrogen stress with the maximum reduction of 53 and 54% in W and NW compared to C at the time of maximum water stress. Genotypic variation of EBv within each treatment was consistently high and over 80%, while the analysis across treatments showed that as stress got higher the genetic variation got lower (32-38%) and the interaction term became more prominent. Phenotypic correlations between EBv and other image-derived traits at early stages showed that under no stress or mild stress conditions, morphological traits were more appropriate than color-related traits for the prediction of biomass accumulation, while under more severe stress conditions, color-related traits and chlorophyll fluorescence are more useful to differentiate genotypes for high biomass. To investigate whether ILs can express EBv in similar patterns, temporal profiles were clustered by using the fuzzy c-means clustering algorithm, and two temporal dynamics of EBv patterns among the studied ILs in each treatment were identified.

Chilling induced stress sRNAs in maize plants in early stages of development.

AB  - Small non-coding RNAs (sRNAs) have an important role in regulating plant gene expression,
both in developmental processes and stress responses, through RNA interference or silencing.
sRNAs respond to environmental stressors in manners dependant on the type of stress, tissue
and genetic background. Herein, their role in chilling stress response is examined in young maize
seedlings (Zea mays L.) of two inbred lines of contrasting chilling tolerance, using high-throughput
sequencing methods. Understanding this response is critical since one of the most promising
strategies of avoiding maize yield loss due to climate change is earlier sowing, during periods
of suboptimal temperatures. Sterilized seeds of the two lines, marked as LT (tolerant), and LS
(sensitive), were germinated in the dark for five days (optimal conditions), after which the 5-d
old seedlings were exposed to chilling conditions for 24h (temperature: 10/8 °C; photoperiod:
12/12h). Samples for RNA isolation were taken after 6h and 24h of treatment. After the cDNA
library preparations were finished, single-end 50 bp sequencing was performed (Illumina® Novaseq 6000). sRNAseq analysis was performed using a custom pipeline that included quality control, mapping, known miRNA alignment and novel miRNA prediction and quantification. This is an ongoing research and a selection of results will be presented. On average 81% of sRNA reads were succesfully mapped and belong to the class of microRNAs (miRNAs). Some of the sRNAs appear to be differentialy regulated depending on the genotype and the chilling conditions, but more research is needed to verify their potential role in this process.

abstract = "Small non-coding RNAs (sRNAs) have an important role in regulating plant gene expression,
both in developmental processes and stress responses, through RNA interference or silencing.
sRNAs respond to environmental stressors in manners dependant on the type of stress, tissue
and genetic background. Herein, their role in chilling stress response is examined in young maize
seedlings (Zea mays L.) of two inbred lines of contrasting chilling tolerance, using high-throughput
sequencing methods. Understanding this response is critical since one of the most promising
strategies of avoiding maize yield loss due to climate change is earlier sowing, during periods
of suboptimal temperatures. Sterilized seeds of the two lines, marked as LT (tolerant), and LS
(sensitive), were germinated in the dark for five days (optimal conditions), after which the 5-d
old seedlings were exposed to chilling conditions for 24h (temperature: 10/8 °C; photoperiod:
12/12h). Samples for RNA isolation were taken after 6h and 24h of treatment. After the cDNA
library preparations were finished, single-end 50 bp sequencing was performed (Illumina® Novaseq 6000). sRNAseq analysis was performed using a custom pipeline that included quality control, mapping, known miRNA alignment and novel miRNA prediction and quantification. This is an ongoing research and a selection of results will be presented. On average 81% of sRNA reads were succesfully mapped and belong to the class of microRNAs (miRNAs). Some of the sRNAs appear to be differentialy regulated depending on the genotype and the chilling conditions, but more research is needed to verify their potential role in this process.",

Nutritional and cost effects of adapted quality protein maize on broiler feeding

AB  - ZPQPM13 is a maize hybrid with improved protein quality, adapted to temperate climate growth conditions, developed by crossing a QPM version of ZPL5 commercial line (developed by marker assisted selection) and GS-6 (a QPM line with 50% tropical and 50% temperate germplasm). This hybrid has grain yield as commercial hybrids, over 90% of hard endosperm, contents of tryptophan and protein over 0.075% and 10%, respectively, and stability of tryptophan content over different locations and years. ZPQPM13 was tested for use in broiler feeds, with the aim to reduce utilization of costly synthetic lysine or soybean components. The feeding experiment on Ross 308 broilers was divided into two groups – control (fed with standard maize) and treatment (fed with QPM). In treatment group diets, ZPQPM13 was increased and soybean component decreased for 3%. Chemical analyses revealed higher contents of limiting essential amino acids in mercantile ZPQPM13 compared to standard maize – 0.44 vs. 0.25 % lysine, 0.077 vs. 0.070 % tryptophan and 0.39 vs. 0.27 % threonine. Duration of each feeding trial was 42 days, comprising three phases - starter (1-14 days), grower (15-35 days) and finisher (36-42 days). Feed intake (FI), body mass gain (BMG), average daily gain (ADG) and feed conversion ratio (FCR) were calculated at the end of each phase. In treatment group, FI was reduced, while BMG and ADG were higher in starter phase but lower in grower and finisher phases. FCR was lower in all three phases in treatment group – 1.13 vs. 1.30, 1.25 vs. 1.33 and 1.28 vs. 1.45. Although a small drawback is that final weight was 3.4 % higher in control broilers, comparison of feed and meat prices show that financial benefits of using QPM are still significant compared to using standard maize in feeds, as soybean is four times more expensive than maize.

period and thus secure maize yields. Earlier sowing dates increase the risk of exposure of the plants to cold during the early development stages
and thus re-evaluation of elite material is required to select more tolerant genotypes. The first step of re-evaluation is to define characteristics that
could be used for fast survey of the elite material, before performing more profound analyses. The aim of this paper was to analyse morphological
characteristics (radicle length, coleoptile length, seedling weight) of 5 day old seedlings of two inbred lines LK and Mo17 (according to breeders
experience tolerant and susceptible, respectively) subjected to chilling stress (10°C day/8°C night) for 24h and 48h, as well as to perform
morphological (root and shoot characteristics) and physiological (chlorophyl, flavonoid and anthocyanin contents) analyses of the plants after
seven day recovery. Control plants were grown under optimal conditions (30°C day /20°C night, ≈60% humidity), at an 8h photoperiod. The
results were not straightforward, as the susceptible Mo17 showed faster recovery (statistically insignificant differences between control and
treatment plants for most of the analysed traits), while the tolerant LK was a more robust genotype throughout the experiment. Chlorophyl,
flavonoid and anthocyanin contents were not informative. The results identified survival rate and root characteristics as informative for fast
screening of seedlings for determination of cold tolerance.

AB  - Finding new ways of improving crop quality, yield potential and abiotic stress tolerance are some of the most important pursuits in crop production today. As one of the biggest causes of yield and productivity reduction is climate change, specifically increasing temperatures and drought during the summer, a large number of strategies is focussed on lessening their negative effects. Cropping pattern changes include earlier sowing (early spring), when the temperatures are lower, as one of the most promising escape strategies for avoiding high summer temperatures. Thus, development of cold tolerant maize lines became an important goal.
Comparative analysis of 46 maize inbred lines belonging to two different genetic backgrounds, one predominantly cold tolerante (marked as Non-Lancaster) and the other predominantly cold sensitive (marked as Lancaster) in the field, was done by whole transriptome sequencing and differential gene expression (DGE) analysis.  Plants were grown under optimal, greenhouse conditions and sampled after completing the V4 growth stage. Total RNA isolated from leaves of three plants per inbred line was used for cDNA library preparation by Illumina TruSeq Stranded RNA LT kit. Pair-end sequencing was performed on MiSeq Illumina sequencer using MiSeq Reagent kit, v2 (2 x 150bp). Data manipulation and analysis was performed using a custom-made bioinformatics pipeline that included high throughput sequence data quality control (using FastQC), removal of low quality reads (using Trimmomatic tool, version 0.32), transcriptome assembly and mapping (using Cufflinks, version 2.2.1), expression quantification (using CuffDiff) and DGE analysis (using BLAST2GO and GO analysis Toolkit and Database for Agricultural Community, agriGO v2).  
DGE analysis revealed 77 differentially expressed genes (DEGs) between the Lancaster and the Non-Lancaster group, 21 of which were  statistically supported for differential expression between the two groups and annotated as involved in abiotic stress responses in maize and other plant species. As one of the biggest causes of yield and productivity reduction is climate change, specifically increasing temperatures and drought during the summer, a large number of strategies is focussed on lessening their negative effects. Thus, development of cold tolerant maize lines became an important goal.
Comparative analysis of 46 maize inbred lines belonging to two different genetic backgrounds, one predominantly cold tolerante (marked as Non-Lancaster) and the other predominantly cold sensitive (marked as Lancaster) in the field, was done by whole transriptome sequencing and differential gene expression (DGE) analysis.  Plants were grown under optimal, greenhouse conditions and sampled after completing the V4 growth stage. Total RNA isolated from leaves of three plants per inbred line was used for cDNA library preparation by Illumina TruSeq Stranded RNA LT kit. Pair-end sequencing was performed on MiSeq Illumina sequencer using MiSeq Reagent kit, v2 (2 x 150bp). Data manipulation and analysis was performed using a custom-made bioinformatics pipeline that included high throughput sequence data quality control (using FastQC), removal of low quality reads (using Trimmomatic tool, version 0.32), transcriptome assembly and mapping (using Cufflinks, version 2.2.1), expression quantification (using CuffDiff) and DGE analysis (using BLAST2GO and GO analysis Toolkit and Database for Agricultural Community, agriGO v2).  
DGE analysis revealed 77 differentially expressed genes (DEGs) between the Lancaster and the Non-Lancaster group, 21 of which were  statistically supported for differential expression between the two groups and annotated as involved in abiotic stress responses in maize and other plant species. Maize is especially sensitive to extreme heat occurring during the flowering and grain filling stages in summer, and earlier sowing enables avoiding this. However, it also means exposure to suboptimal temperatures and chilling stress during earlier developmental stages, leading to a demand for the development of maize lines tolerant to low temperatures during those stages.
In this study, 46 maize lines used in breeding programmes were grouped as Lancaster and group consisting of different heterotic groups like BSSS, Iowa dent, etc. and further studied by whole transcriptome sequencing (maize leaves, V4 stage, optimal temperature conditions). Gene expression analyses revealed a set of 77 differentially expressed genes (DEGs) between the two groups, out of which 20 were annotated as related to abiotic stress response. ATP synthase CF0 A subunit gene (atpI) was chosen for further characterization under low temperature conditions in two inbred lines (L1, L2) with most contrasting FPKM values, one belonging to each group. The experiment was performed with 5-day old and V4 maize seedlings, under optimal (25°/20°C) and low (8°/10°C) temperature conditions, with a 12h photoperiod. Samples for RNA extraction, cDNA synthesis and qPCR expression analysis were taken after 6h and 24h exposure to experimental temperatures. 
The results showed different expression regulation of atpI dependent on cold exposure duration, developmental stage and genetic background. atpI expression was up-regulated in both genotypes in V4 stage, with the expression peak after 6h of treatment. In the 5-day old seedling stage, atpI expression depended on the genotype – it was down-regulated in L1, and up-regulated in L2. The expression in both genotypes in this developmental stage was at its highest after 24h of treatment. This suggests that mechanisms involved in ATP synthesis and photosynthetic phosphorylation are differentially regulated based on low temperature exposure duration, developmental stage and genetic background.

Sowing maize in early spring, when the temperatures are lower, enables avoidance of drought and high summer temperatures during the flowering and grain filling stages, but it also means that maize plants will be exposed to suboptimal temperatures during earlier developmental stages. Consequently, development of maize lines tolerant to low temperatures during those stages becomes precedence. 
An initial study encompassed whole transcriptome sequencing of 46 maize inbred lines at V4 stage grown under optimal temperature conditions. Gene expression analysis of maize genotypes grouped as Lancaster and non-Lancaster (BSSS, Iowa dent, etc.) revealed a set of 77 differentially expressed genes (DEGs). Their expression showed specific profiles depending on the duration of low temperature exposure and genetic background. To test if these genes follow the same expression patterns in earlier developmental stages, the experiment was performed with 5-day old maize seedlings of two inbred lines (tolerant and sensitive), under optimal (25°/20°C) and low (8°/10°C) temperature conditions, with a 12h photoperiod. Samples for RNA extraction, cDNA synthesis and qPCR expression analysis were taken after 6h and 24h exposure to experimental temperatures. Five analyzed genes showed different expression regulation dependent on cold exposure duration. Two genes showed regulation dependent both on cold exposure duration and genetic background. Additionally, three of five DEGs showed different expression patterns at 5-day old seedling stage than at V4 stage. The results imply that processes underlying maize low temperature response are differentially regulated based on stress exposure duration, developmental stage and genetic background.

One of those strategies is earlier sowing, during the colder months in early spring, which means that finding methods that quickly and accurately estimate low temperature tolerance of different crops, including maize, is becoming increasingly important. Our idea was to use the expression of Zea mays sp. cold induced genes (ZmCOI) for this purpose, and to see if there is a significant correlation between the quantities of certain ZmCOI mRNA and low temperature tolerance. This was done through an experiment that included growing several maize lines known from breeders experience to be sensitive or tolerant to cold, under optimal and low temperatures, 22° C and 6° C, respectively. Samples (plant leaves) were taken during the V4 growth stage, at different time points during the treatment (6h and 24h) and used for RNA extraction, cDNA synthesis and qPCR expression analysis for the selected genes (ZmDREB2A, ZmERF3, ZmACA1). Preliminary results showed changes in the expression of these genes between chosen sensitive and tolerant inbred lines. These results provide a good basis for further studies with more cold induced genes and inbred lines that will hopefully confirm their effectiveness as a tool for determination of tolerance to low temperatures.

AB  - One of the strategies for overcoming global climate change threatening to decrease maize yield is early sowing. To contribute to the development of cold-tolerant hybrids this research focused on the genetic background's comparative analysis in maize inbreds with good combining ability. Leaf whole-transcriptome sequencing of 46 maize genotypes revealed 77 differentially expressed genes (DEGs) between Lancaster and other heterotic groups (i.e. BSSS, Iowa dent, Ohio), referred to as non-Lancaster group, under optimal growing conditions. Cold test of the subset of four Lancaster and four non-Lancaster lines showed that the former were cold sensitive and the latter cold tolerant. Cold-induced expression analysis of seven DEGs in eight lines revealed different expression regulation dependent on the duration of cold exposure and genetic background for six out of seven analysed genes—chloroplast ATP-sulphurylase, photosystem II cytochrome b559 alpha subunit, CIPK serine-threonine protein kinase 15, glutamyl-tRNA reductase, photosystem II reaction centre protein I and Calvin cycle CP12-chloroplastic-like encoding genes. The results imply that differently regulated basic processes between Lancaster and non-Lancaster maize group involve, at least, photosynthesis and sulphate assimilation, contributing to their different cold response and different adaptation to low temperatures.

Adapted quality protein maize for broiler feeds

AB  - Maize has low nutritional quality due to poor content of essential amino
acids lysine and tryptophan in the dominant seed storage protein fraction,
zeins. However, Quality Protein Maize (QPM), a variety of opaque2 maize
but with good agronomic traits, can have 60% to 100% higher content of
lysine than standard maize. Monogastric animals (pigs, poultry, fish), like
humans, cannot synthesize lysine and tryptophan de novo and thus these
amino acids must be supplied through diets. QPM was primarily developed
for human consumption to overcome malnutrition in countries where maize
is staple food. Thus, QPM is of tropical origin and its adaptation to temperate
regions is frequently hampered by the retained exotic germplasm.
Development of adapted QPM hybrids is mainly aimed for feed industry. It
has been shown that substituting standard maize with QPM in feed diets
could be profitable due to improved weight gain, feed conversion ratio and
decreasing of dietary lysine supplementation. Maize Research Institute,
Zemun Polje has a program on converting elite inbred lines through marker
assisted breeding into their QPM counterparts and developing commercial
QPM hybrids. Up to now, two inbred lines have been converted, while eight
lines have passed through three or four generations of backcrossing and one
generation of selfing. Tryptophan content in the converted selfed plants was
in the range from 0.080 to 0.093. Besides developing QPM counterparts of
commercial hybrids, one QPM hybrid obtained by crossing a converted elite
inbred line and an adapted tropical inbred line was developed and tested over
different locations and in different years. This hybrid is currently used in
feeding experiments with the objective to test effects of replacing standard
maize with QPM in diets on broiler performances.

AB  - Maize is one of the most important crop species worldwide, but also extremely
susceptible to the effects of increasingly higher temperatures and drought during the
summer and its flowering and grain filling stage. Different strategies are being utilized to
ensure a satisfying yield potential and quality even in the extremely unfavourable
environmental conditions, which are the result of climate change. Some of them are
cropping pattern changes and sowing alterations, including earlier sowing. Since this
implies the exposure to suboptimal temperatures during early developmental stages, it
leads to a demand for developing maize lines tolerant to low temperatures during these
stages. This research focuses on the first phase of germination, imbibition.
Maize tolerance to low temperatures is a complex trait that includes different mechanisms
and strategies that all work together to ensure adaptation and survival, such as cell
membrane changes, antioxidative system activation, etc. Additionally, the low
temperature response of different maize inbreds varies substantially, so recognizing the
different ways in which they respond to low temperatures during imbibition and other
earlier stages of development is crucial. To accomplish this, seeds of two maize inbred
lines of contrasting susceptibility to low temperatures were selected and exposed to
control (20° C) and low temperature (8°C) conditions during the first 24h of imbition andthen further analyzed to assess their response. This included germination and tetrazolium
assays; ascertaining membrane integrity by evaluating cell leakage and lipid peroxidation;
determining the antioxidative capacity by assessing superoxide dismutase (SOD) and
catalase (CAT) activity; and expression analysis of four genes included in the low
temperature response (gibberellin insensitive dwarf 1 gibberellin receptor, gid1; fatty acid
desaturases 2 and 6, fad2 and fad6; plastid-lipid-associated 2 protein, pap2).
The results showed that, while there is not a significant difference in their germination
rate, they differ in their survival rate, with more seeds of the tolerant genotype surviving
the low temperature period. Significant differences between them were found in cell
leakage (p<0,01), as well as gid1(p<0,05) and fad6 (p<0,05) gene expression assays. The
present research brings light to our understanding of the effect of low temperatures on the
first germination stage, - imbibition. It highlights the importance of choosing the right
inbreds for earlier sowing and points to certain routes that could be taken for improving
and accelerating the breeding process for low temperature tolerance.
AB  - Kukuruz je usev od izuzetne važnosti za poljoprivredu, koji se takođe odlikuje značajnom
osetljivošću na visoke temperature i sušu tokom letnjih meseci, odnosno tokom faze cvetanja i
nalivanja zrna. Jedna od strategija obezbeđivanja zadovoljavajuće produktivnosti i prinosa
kukuruza, uprkos nepovoljnim uslovima životne sredine, jeste ranija setva. Ipak u tom slučaju su
biljke kukuruza izložene suboptimalnim temperaturama u ranim fazama razvića, što stvara
potrebu za razvojem linija tolerantnih na niske temperature u tim fazama, što je i fokus ovog
istraživanja. Odgovor kukuruza na niske temperature je kompleksan i uključuje niz različitih
mehanizama i strategija koje obezbeđuju opstanak, a takođe zavisi i od samog genotipa, faze
razvića, itd. Iz tog razloga, odgovor kukuruza na niske temperature (8°C) je posmatran tokom
faze imbibicije, kod dva genotipa, i to na nivou promena genske ekspresije (gid1; fad2; fad6;
pap2), aktivnosti antioksidativnih enzima (SOD, CAT), integriteta ćelijskih membrana, stopa
germinacije, kao i samog preživljavanja. Rezultati ukazuju da postoje statistički značajne razlike
između genotipova kada je reč o stopi preživljavanja, integritetu ćelijskih membrana i ekspresiji
gena gid1i fad6 (p<0,05). Ovo istraživanje ukazuje na moguće mehanizme aklimatizacije na
niske temperature tokom faze imbibicije i ističe značaj odabira odgovarajućih linija za raniju
setvu.

inbreds for earlier sowing and points to certain routes that could be taken for improving
and accelerating the breeding process for low temperature tolerance., Kukuruz je usev od izuzetne važnosti za poljoprivredu, koji se takođe odlikuje značajnom
osetljivošću na visoke temperature i sušu tokom letnjih meseci, odnosno tokom faze cvetanja i
nalivanja zrna. Jedna od strategija obezbeđivanja zadovoljavajuće produktivnosti i prinosa
kukuruza, uprkos nepovoljnim uslovima životne sredine, jeste ranija setva. Ipak u tom slučaju su
biljke kukuruza izložene suboptimalnim temperaturama u ranim fazama razvića, što stvara
potrebu za razvojem linija tolerantnih na niske temperature u tim fazama, što je i fokus ovog
istraživanja. Odgovor kukuruza na niske temperature je kompleksan i uključuje niz različitih
mehanizama i strategija koje obezbeđuju opstanak, a takođe zavisi i od samog genotipa, faze
razvića, itd. Iz tog razloga, odgovor kukuruza na niske temperature (8°C) je posmatran tokom
faze imbibicije, kod dva genotipa, i to na nivou promena genske ekspresije (gid1; fad2; fad6;
pap2), aktivnosti antioksidativnih enzima (SOD, CAT), integriteta ćelijskih membrana, stopa
germinacije, kao i samog preživljavanja. Rezultati ukazuju da postoje statistički značajne razlike
između genotipova kada je reč o stopi preživljavanja, integritetu ćelijskih membrana i ekspresiji
gena gid1i fad6 (p<0,05). Ovo istraživanje ukazuje na moguće mehanizme aklimatizacije na
niske temperature tokom faze imbibicije i ističe značaj odabira odgovarajućih linija za raniju
setvu.",

AB  - Nutritional  quality  of  maize  is  low  because  maize  protein  is  poor  in  several  essential amino  acids.  The  purpose  of  this  research  was  to  analyze  agronomic  traits  and  kernel biochemical and physical properties of 16 gene bank accessions which comprise a mini-core  collection  for  grain  quality  and  to  identify  populations  for  improving  protein quality.  Standard  ZP341  hybrid  was  superior  for  half  of  agronomic  traits  tested, especially  grain  yield,  which  was  higher  from  24%  to  six  times.  Ten  accessions  had protein  content  over  14  %  and  were  further  analyzed  for  amino  acid  composition  and kernel  characteristics.  Additionally,  genetic  relationships  between  the  accessions  were determined   by   Simple   Sequence   Repeats   (SSRs)   analysis   with   30   primers.   All accessions  showed  elevated  contents  of  most  essential  amino  acids.  Population  L492 with  1.87  and  0.68  g  100g-1dry  weight  had  the  highest  contents  of  leucine  and phenylalanine, respectively, but also higher contents of most other analyzed amino acids (p<0.05). Cluster analysis based on SSRs also distinguished L492 by separating it from all other accessions. Compared to ZP341, accessions were significantly inferior in grain weight  and  dimensions  (p<0.05),  but  superior  in  most  hardness  parameters  (p<0.05). Pearson  correlations  revealed  lack  of  negative  correlations  between  biochemical  traits, indicating  a  possibility  for  concurrent  improvement  of  several  amino  acids.  The  best way  of  improving  protein  quality  of  elite  materials  is  through  backcrossing  and  as populations  were  chosen  according  to  their  good  general  combining  ability  (with IoDent,  Lancaster  and  BSSS),  they  could  serve  for  improvement  of  elite  materials  of these genetic origins.

Svi  uzorci  su  pokazali  povećan  sadržaj  većine  esencijalnih  amino  kiselina. Populacija L492 je imala najveći sadržaj leucina (1.87g 100g-1suve  mase)  i  fenilalanina  (0.68g 100g-1suve  mase),  ali  i  veće  sadržaje  ostalih  aminokiselina  (p<0.05)  u  odnosu  na  ZP341  i analizirane  populacije.  Klaster  analiza  zasnovana  na  SSR  markerima  je  takoĊe  izdvojila populaciju  L492  od  svih  ostalih  populacija.  U  odnosu  na  ZP  341,  populacije  iz  banke  gena  su bile  inferiorne  u  masi  i  dimenzijama  zrna  (p<0.05), ali superiorne u većini parametara tvrdoće zrna   (p<0.05).  Pirsonove   korelacije  su  pokazale  nedostatak  negativnih  korelacija  izmeĊu analiziranih  biohemijskih  svojstava,  što  ukazuje  na  mogućnost  poboljšanja  kukuruza  na  više amino kiselina istovremeno. Najbolji naĉin poboljšanja kvaliteta proteina elitnog materijala je putem   povratnih  ukrštanja,  a  kako  su  populacije  izabrane  prema  svojim  dobrim  opštim kombinacionim sposobnostima (sa IoDent, Lancaster i BSSS), mogle bi da služe za poboljšanje elitnog materijala navedenih heterotiĉnih grupa.

AB  - In this paper agronomic characteristics of quality protein maize (QPM) hybrids adapted to temperate
regions created in Maize Research Institut Zemun Polje are presented. Hybrid ZPQPM13 showed
favorable traits for growing in these regions, and its grain yield was at the level of one of the leading
ZP hybrids - ZP 606. However, grain moisture of this hybrid was 3% higher than that of ZP 606,
pointing out to the tropical origin of its parental inbred lines. Namely, mother was obtained from F1
cross between tropical and adapted lines, and father by marker assisted selection (MAS) from QPM
line of tropical origin that was crossed and twice backcrossed with a commercial ZP line. In the future,
in order to obtain even more adapted QPM hybrids, MAS program will continue with all the best
commercial ZP inbred lines of all FAO groups.

where maize is the main source of protein. Although its cultivation in temperate regions is hampered by residues of
exotic germplasm, it could be beneficial for use in livestock feeds as it was shown that substitution of standard maize
with QPM can improve livestock characteristics and decrease dietary lysine supplementation. The aim of this study
was to test 11 QPM hybrids obtained by crossing adapted QPM inbred lines for their performance in field trials in 2
yr at four locations, as well as to determine their relevant biochemical components. The main reason for rejecting nine
QPM hybrids was low grain yield, standard hybrids had higher yields on average for 37.8%. Hybrid ZPQPM6 had good
agronomic characteristics, but its biochemical components were nonsignificantly different from the standard hybrid.
Only hybrid ZPQPM13 met necessary criteria, grain yield comparable with standard hybrids, high tryptophan content
in different environments (average 0.083%) and hard endosperm (average score 1.87). Lysine content, measured after
mercantile production, was 0.44%. Quality index, although below the QPM threshold (which is 0.80%) was significantly
higher (p < 0.05) in ZPQPM13 in comparison with standard hybrid, indicating improved nutritional quality of the protein.
The results indicated that presence of exotic germplasm in these QPM hybrids is a consequential difficulty and that in their
parental inbred lines at least one more backcross with temperate germplasm should be done to select better adapted QPM.

used to avoid drought during flowering and diminish yield losses. Herein, a small-scale
experiment of low temperature stress (LTS) on maize lines L1 (tolerant), L2 (medium
tolerant) and L3 (susceptible) is presented. Plants were grown in pots exposed to
exterior suboptimal (March) and optimal (late April) temperatures until three leaf stage.
Chlorophyll (CH), flavonoids (FL), anthocyanins (AN) and nitrogen balance (NBI)
indices were measured using Dualex Scientific optical device. Growth parameters were
also determined. Under LTS, number of plants was unchanged for L1 and halved for L2
and L3. Compared to L2 and L3, L1 had significantly higher (p<0.05) shoot fresh
weight (0.649 g vs. 0.406 g and 0.303 g), AN (0.17 vs. 0.13) and FL (1.47 vs. 1.38 and
1.36). For recovery evaluation, plants were transplanted into the field. Transplanted
stressed L1 plants showed the highest grain yield per plant (55g) in the field. Due to
high correlations (p<0.01) between FL in three leaf stage and grain yield per plant, FL
could be used as an indicator of plant recovery of maize genotypes exposed to LTS
during early sowing.
AB  - Ranom setvom kukuruza se može izbeći efekat suše u fazi cvetanja i tako preduprediti smanjenje
prinosa uprkos nepovoljnim temperaturama u tom periodu. U ovom istraživanju prezentovani su
rezultati efekta niskih temperatura na tri linije kukuruza: L1 (tolerantna), L2 (srednje osetljiva) i
L3 (osetljiva). Biljke su gajene do faze trećeg lista u saksijama izloženim suboptimalnim (mart) i
optimalnim (april) spoljnim temperaturama. Sadržaj hlorofila, flavonoida i antocijana kao i
nitrogen balance index (NBI) su mereni korišćenjem uređaja Dualex Scientific (Force-A, Orsay,
France). Takođe, mereni su i parametri rasta. U poređenju sa L2 i L3, L1 je imao značajno veću
(p<0.05) svežu masu nadzemnog dela biljke (0.649 g vs. 0.406 g i 0.303 g), antocijana (0.17 vs.
0.13) i flavonoida (1.47 vs. 1.38 i 1.36). Biljke su presađene u polje radi procene oporavka.
Presađene biljke genotipa L1 su pokazale najveći prinos po biljci u polju (55g). S obzirom na
visoku korelaciju između sadržaja flavonoida u fazi trećeg lista i prinosa po biljci (p<0.01),
flavonoidi mogu biti korišćeni kao indikator oporavka biljke kukuruza izloženih niskim
temperaturama u fazi rane setve

Marker assisted selection for β-carotene rich maize: two-level selection procedure in BC2 generation

AB  - This paper is a part of the breeding program conducted at the Maize Research Institute "Zemun Polje", with the aim to convert the standard maize to β-carotene rich genotypes adapted to temperate regions using the marker assisted selection. When SSR marker specific to the crtRB1 gene has been tested on parental lines, distinct polymorphism was observed between the donor and recurrent parents. The same marker has also been validated in BC1 generation when heterozygous individuals were clearly distinguishable from the homozygous dominants. The results presented herein refer to two-level selection procedure in BC2 generation. First, BC2 plants were analyzed with crtRB1-specific molecular marker to separate heterozygotes from dominant homozygotes. Percentage of heterozygous plants was approximately 50%, which was in accordance with the expected Mendelian ratio of 1:1. Second, the selected heterozygotes were screened with 30 polymorphic SSR markers distributed throughout the maize genome to identify genotypes with the highest recovery of recurrent parent’s genome (RPG). The RPG values among three analyzed parental lines and their respective progenies ranged from 85-99%. For each line separately, RPG values were: 86-97% (RP1), 90-95% (RP2) and 85-99% (RP3). Plants with RPG above 95% were selfed to produce BC2F2 generation in which homozygous recessive individuals would be identified. Those genotypes will be subjected to biochemical and phenotypic evaluation to confirm their nutritional and agronomical superiority. Finally, these β-carotene enriched lines, as well as the resulting improved hybrids, can be used in the biofortification programs.
