AB  - This work aimed to evaluate changes in phytic phosphorus content as the response of three
maize hybrids (ZP 457, ZP 5601, and ZP 606) to different pesticide treatments for European Corn
Borer (ECB) attacks and, as well as the correlation between damage caused by ECB attack and
Pphy content. In the experimental field, maize ears were sampled and dried at a moisture content
of 14% for the determination of changes in phytic phosphorus content in the grain. The content of phytic phosphorus was determined using UV / VIS spectrophotometry. Observed results showed the highest content of phytic phosphorus for untreated control for all tested hybrids, compared to pesticide treatments. The content of Pphy in the tested maize kernel samples ranged from 2.12 to 3.40 mg g -1. A significant positive correlation between attacks of larvae and Pphy content could indicate activation of its antioxidative function as a response to damage caused by ECB attack.
AB  - Cilj ovog rada je procena promene u sadržaju fitinskog fosfora kao odgovora tri hibrida kukuruza (ZP 457, ZP 5601 i ZP 606) na različite tretmane pesticidima protiv napada kukuruznog
plamenca, kao i određivanje korelacije između oštećenja biljke izazvane napadom larvi i sadržaja
fitinskog fosfora. U oglednom polju uzorkovani su klipovi kukuruza radi određivanja promene
sadržaja fitinskog fosfora u zrnu UV/VIS spektrofotometrijom. Dobijeni rezultati su pokazali najveći sadržaj fitinskog fosfora za netretiranu kontrolu kod svih ispitivanih hibrida u poređenju sa
tretmanima pesticidima. Sadržaj fitinskog fosfora u ispitivanim uzorcima zrna kukuruza kretao
se od 2,12 do 3,40 mg g-1. Uočena značajna pozitivna korelacija između napada larvi i sadržaja fitinskog P može ukazivati na aktivaciju njegove antioksidativne funkcije kao odgovor na oštećenja izazvana napadom kukuruznog plamenca.

MARKER-ASSISTED GА1-S INCORPORATION INTO WHITE MAIZE HYBRID´S PARENTAL LINES

AB  - Maize Research Institute Zemun Polje has a marker assisted backcross breeding (MABB) program aimed to incorporate the strong allele of incompatibility gene Gа1 into parental components of the hybrids with specific traits (white kernel). The main objective of this research was marker assisted development of the white maize hybrid´s parental lines with the complete cross-incompatibility to foreign pollens. Ga1-S-tightly-linked molecular markers successfully identified heterozygous plants with the percentage of approximately 50% after all three backcrossing. Regarding the background selection, 69% of progenies had recurrent parent’s genome (RPG) above the theoretical value for BC3 generation. Also, a few individuals even had 99%, the value theoretically achieved in BC6 generation, showing that MABB made a genetic gain in RPG recovery. Foreground selection in this generation aimed to identify homozygous dominant individuals. Out of 264 BC3F2 plants, 70 (26.5%) were dominant homozygotes, which is in accordance with the expected Mendelian ratio. Progenies of the BC3F2 homozygous dominant plants were planted alternatively with yellow-kernel maize of the same maturity to check cross-incompatibility. Unfortunately, none of the dominant progenies had a 100% white kernel, most likely due to the presence of modifier genes that increase/decrease the effectiveness of pollen exclusion or that, alternatively, modifiers were lost during the backcrossing. It could be expected that successfully employed functional markers (developed from the sequence polymorphisms present within functional gene(s) associated with phenotypic trait variations) would outbalance the noted impediments and enhance MABB efficiency to transfer the desired gene(s) controlling simple or complex trait(s) into cultivated varieties.
AB  - U Institutu za kukuruz “Zemun Polje” postoji program selekcije pomoću molekularnih markera (MAS) čiji je cilj ugradnja gena za inkompatibilnost Gа1-S u linije kukuruza koje su roditeljske komponente hibrida belog zrna. Glavni cilj ovog rada je bio stvaranje linija kukuruza belog
zrna sa potpunom unakrsnom inkompatibilnošću sa stranim polenom pomoću molekularnih
makera. Molekularnim markerima koji su blisko vezani za Ga1-S gen uspešno su identifikovane
heterozigotne biljke (50%) nakon sva tri povratna ukrštanja. Rezultati utvrđivanja procenta genoma rekurentnog roditelja (RPG) u BC3 generaciji pokazali su vrednost iznad očekivane kod 69%
potomstva, dok je kod određenog broja RPG iznosio 99%, što predstavlja vrednost koja se teoretski dostiže u BC6 generaciji. Od 264 BC3
F2 pojedinačnih biljaka, 70 (26,5%) je bilo dominantno
homozigotno, što je u skladu sa očekivanim, prema pravilima Mendelovog nasleđivanja. Potomstva BC3
F2 dominantno homozigotnih biljaka posejana su sa kukuruzom žutog zrna iste grupe
zrenja radi provere unakrsne inkompatibilnosti. Nijedno od dobijenih potomstava nije imalo 100%
belo zrno, najverovatnije usled prisustva gena modifikatora koji menjaju efikasnost isključivanja
polena, ili je došlo do gubljenja ovih gena prilikom povratnog ukrštanja. Moglo bi se očekivati da
će uspešna primena funkcionalnih markera (stvorenih na osnovu polimorfizama sekvenci unutar
samog gena vezanog za fenotipsku osobinu) prevazići uočene prepreke i poboljšati efikasnost MAS
za prenos željenih gena koji kontrolišu prostu ili složenu osobinu u gajene kulture.

Tryptophan content increase in maize inbred lines improved through marker assisted breeding

AB  - Quality protein maize (QPM) has high content of essential amino acids lysine and tryptophan that are deficient in standard maize. Naturally occurring opaque2 recessive mutation has been the most widely studied and used as a source for genetic improvement of the nutritional value of maize proteins. Marker assisted conversion of maize inbred lines to QPM adapted to temperate regions is being conducted at Maize Research Institute Zemun Polje (MRIZP). The ultimate goal is selection of the opaque2 recessive genotypes (foreground selection) with the highest proportion of recurrent parentʹs genome (background selection) along with high lysine/tryptophan content without losing good agronomic performances of the original line. After two backcrossing and selection of heterozygous individual plants, opaque2-specific molecular markers phi057 and umc1066 successfully identified recessive homozygotes in BC2F2 generation. The next step was to confirm expression of the QPM trait through elevated kernel tryptophan content (TC). Laboratory analyses were used to quantify and select for acceptable tryptophan concentrations for QPM - above 0.075%. The results presented in this paper relate to identification of BC2F3 progenies with increased TC in the conversion of two MRIZP commercial inbred lines (RP1 and RP2). The average TC ranged from 0.070% in RP1 to 0.074% in RP2. Out of 60 progenies from these two lines, 27 had TC at or above the QPM treshold. A total of 12 progenies was chosen for the highest tryptophan content. These lines will serve as parental lines for developing QPM hybrids adapted to temperate regions.

Morphological diversity of soybean in Maize Research Institute collection

AB  - Maize Research Institute Zemun Polje soybean collection is a small, local collection consisted of
approximately 500 accessions from different regions of the world. Until now, there has been no
systematic and precise evaluation of samples in the collection, as well as the assessment of
genetic diversity. Genotype’s information is mainly limited to passport data, a description of
several morphological traits and yield data. The aim of this study was to evaluate the level of
phenotypic variation in collection on the sample of 90 accessions of different geographical origin.
Morphological description was performed by screening of 16 traits according to the UPOV
technical guideline for conducting of DUS tests in soybean. Diversity of morphological traits was
estimated by Shannon diversity index. Morphological similarity of genotypes was estimated by the
application of multivariate statistics models (analysis of homogeneity - HOMALS and cluster 
analysis). HOMALS analysis revealed the descriptors of high discriminatory power, with the 
greatest importance for genotype’s distinction: colour of the seed coat, intensity of pod colour,
pubescence colour, and to a lesser extent the growth type. Genotypes were divided into six
homogeneous and moderately separated groups with similar morphological profiles. Geographical
component of morphological variability, as well as pedigree-based grouping could not be observed
in pattern obtained by HOMALS. Cluster analysis showed a slightly different grouping model
compared to the HOMALS analysis. All genotypes were grouped according to the colour of flower
into two main clusters, and further into subgroups according to the type of growth and
pubescence colour. Genotype’s positioning in subclusters did not correspond to their geographical

approximately 500 accessions from different regions of the world. Until now, there has been no
systematic and precise evaluation of samples in the collection, as well as the assessment of
genetic diversity. Genotype’s information is mainly limited to passport data, a description of
several morphological traits and yield data. The aim of this study was to evaluate the level of
phenotypic variation in collection on the sample of 90 accessions of different geographical origin.
Morphological description was performed by screening of 16 traits according to the UPOV
technical guideline for conducting of DUS tests in soybean. Diversity of morphological traits was
estimated by Shannon diversity index. Morphological similarity of genotypes was estimated by the
application of multivariate statistics models (analysis of homogeneity - HOMALS and cluster 
analysis). HOMALS analysis revealed the descriptors of high discriminatory power, with the 
greatest importance for genotype’s distinction: colour of the seed coat, intensity of pod colour,
pubescence colour, and to a lesser extent the growth type. Genotypes were divided into six
homogeneous and moderately separated groups with similar morphological profiles. Geographical
component of morphological variability, as well as pedigree-based grouping could not be observed
in pattern obtained by HOMALS. Cluster analysis showed a slightly different grouping model
compared to the HOMALS analysis. All genotypes were grouped according to the colour of flower
into two main clusters, and further into subgroups according to the type of growth and
pubescence colour. Genotype’s positioning in subclusters did not correspond to their geographical

AB  - U Institutu za kukuruz “Zemun Polje” postoji program selekcije pomoću molekularnih markera (MAS) čiji je cilj ugradnja gena za inkompatibilnost Gа1-S u linije kukuruza koje su roditeljske komponente hibrida belog zrna. Glavni cilj ovog rada je bio stvaranje linija kukuruza belog zrna sa potpunom cross-inkompatibilnošću sa stranim polenom pomoću molekularnih makera. Molekularnim markerima koji su blisko vezani za Ga1-S gen uspešno su identifikovane heterozigotne biljke (50%) nakon sva tri povratna ukrštanja. Rezultati utvrđivanja procenta genoma rekurentnog roditelja (RPG) u BC3 generaciji pokazali su vrednost iznad očekivane kod 69% potomstva, dok je kod određenog broja RPG iznosio 99% što predstavlja vrednost koja se teoretski dostiže u BC6 generaciji. Od 264 pojedinačne biljke BC3F2 generacije, 70 (26,5%) su bili dominantni homozigoti, što je u skladu sa očekivanim prema pravilima Mendelovog nasleđivanja. Potomstva BC3F2 dominantno homozigotnih biljaka posejana su sa kukuruzom žutog zrna iste grupe zrenja radi provere cross-inkompatibilnosti. Nijedno od dobijenih potomstava nije imalo 100% belo zrno, najverovatnije usled prisustva gena modifikatora koji menjaju efikasnost isključivanja polena, ili je došlo do gubljenja ovih gena prilikom povratnog ukrštanja. Moglo bi se očekivati da bi se primenom funkcionalnih markera (stvorenih na osnovu polimorfizama sekvenci unutar samog gena vezanog za fenotipsku osobinu) uspešno prevazišle uočene prepreke i poboljšala efikasnost MAS za prenos željenih gena koji kontrolišu prostu ili složenu osobinu u gajene kulture.
AB  - Maize Research Institute Zemun Polje has a marker assisted backcross breeding (MABB) program aimed to incorporate the strong allele of incompatibility gene Gа1 into parental components of the hybrids with specific traits (white kernel). The main objective of this research was marker assisted development of the white maize hybrid´s parental lines with the complete cross-incompatibility to foreign pollens. Ga1-S-tightly-linked molecular markers successfully identified heterozygous plants with the percentage of approximately 50% after all three backcrossing. Regarding the background selection, 69% of progenies had recurrent parent’s genome (RPG) above the theoretical value for BC3 generation. Also, a few individuals even had 99%, the value theoretically achieved in BC6 generation, showing that MABB made a genetic gain in RPG recovery. Foreground selection in this generation aimed to identify homozygous dominant individuals. Out of 264 BC3F2 plants, 70 (26.5%) were dominant homozygotes, which is in accordance with the expected Mendelian ratio. Progenies of the BC3F2 homozygous dominant plants were planted alternatively with yellow-kernel maize of the same maturity to check cross-incompatibility. Unfortunately, none of the dominant progenies had a 100% white kernel, most likely due to the presence of modifier genes that affect the effectiveness of pollen exclusion or that, alternatively, modifiers were lost during the backcrossing. It could be expected that successfully employed functional markers (developed from the sequence polymorphisms present within functional gene(s) associated with phenotypic trait variations) would outbalance the noted impediments and enhance MABB efficiency to transfer the desired gene(s) controlling simple or complex trait(s) into cultivated varieties.

AB  - Upotreba kukuruza u ishrani ljudi i životinja ima široku primenu jer predstavlja
izvor različitih tohemikalija koje povoljno utiču na metaboličke procese. Tokom
vegetacije biljka kukuruza izložena je različitim uticajima abiotičkog i biotičkog
stresa. Napad jedne od najznačajnijih štetočina kukuruza, larve Ostrinia nubilalis
zavisi od uslova sredine i može dovesti do smenjenja prinosa i do 30%. Oštećenja
nadzemnih delova biljke povećavaju rizik od pojave truleži klipa, jer predstavljaju
mesta

sekundarne infekcije izazvane različitim patogenim gljivama. Jedna
od neophodnih agrotehničkih mera je primena pesticida u poljoprivredi u cilju
suzbijanju kako štetočina tako i korova, obezbeđujući stabilan prinosa. S druge
strane, primena pesticida, nepovoljni uslovi spoljašnje sredine i biotički stres mogu
dovesti do oksidativnog stresa, oštećenja ćelija i pojave slobodnih radikala i reaktivnih
vrsta
kiseonika
(engl.
reactive
oxygen
species
-
ROS).
Ćelije
i
tkiva
biljaka

imaju
razvijene
antioksidativne
sisteme
odbrane
koji
ihibiraju
ili
vezuju
slobodne

radikale.

Najznačajniji neenzimski antioksidansi su avonoidi, fenolne kiseline,
karotenoidi, tokoferoli, tinska kiselina, vitamin C, i glutation. Osim promena u 
prinosu i antioksidativnom statusu, kod biljke može doći i do promena u količini
i drugih tohemikalija kao što su proteini, šećeri, amnokiseline i masne kiseline.
Do sada su nedovljno ispitani uticaji pesticida i abiotički stres koji mogu da izazovu
promene
na
različitim
tkivima
kukuruza,
pri
čemu
je
više
pažnje
posvećano

promeni
tohemikalija

izazvanoj uticajem spoljašnje sredine kao što je vodni
decit. U cilju pronalaženja karakterističnog odgovora genotipa, u odnosu na
promene tohemijskog sadržaja, kao i eventualno izdvajanje genotipa tolerantnog
na potencijalni stres primenjuju su metode spekrofotometrije i savremene metode
tečne i gasne hromatograje za odredjivanje tohemikalija u biljci.

AB  - U Institutu za kukuruz "Zemun Polje" postoji program selekcije pomoću molekularnih markera (MAS) čiji je cili ugradnja gena za inkompatibilnost Ga1-S u linije kukuruza koje su roditeljske komponente hibrida belog zrna. Glavni cilj ovog rada je bio stvaranje linija kukuruza belog zrna sa potpunom cross-inkompatibilnošću sa stranim polenom pomoću molekularnih makera. Molekularnim markerima koji su blisko vezani za Ga1-S gen uspešno su identifikovane heterozigotne bilike (50%) nakon sva tri povratna ukrštanja. Rezultati utvrđivanja procenta genoma rekurentnog roditelia (RPG) u BC3 generaciji pokazali su vrednost iznad očekivane kod 69% potomstva, dok je kod određenog broja RPG iznosio 99% što predstavlja vrednost koja se teoretski dostiže u BC6 generaciji. Od 264 pojedinačne bilike BC3F2 generacije, 70 (26,5%) su bili dominantni homozigoti, što je u skladu sa očekivanim prema pravilima Mendelovog nasleđivanja. Potomstva BC3F2, dominantno homozigotnih biljaka posejana su sa kukuruzom žutog zrna iste grupe zrenja radi provere cross-inkompatibilnosti. Nijedno od dobijenih potomstava nije imalo 100% belo zrno, najverovatnije usled prisustva gena modifikatora koji menjaju efikasnost isključivanja polena, ili je došlo do gublienja ovih gena prilikom povratnog ukrštanja. Moglo bi se očekivati da bi se primenom funkcionalnih markera (stvorenih na osnovu polimorfizama sekvenci unutar samog gena vezanog za fenotipsku osobinu) uspešno prevazišle uočene prepreke i poboljšala efikasnost MAS za prenos željenih gena koji kontrolišu prostu ili složenu osobinu u gajene kulture.

AB  - A high level of genetic purity in crop varieties must be achieved and maintained for
agronomic performance, encouraging investment and innovation in plant breeding and ensuring that
the improvements in productivity and quality imparted by breeders are delivered to the consumer.
Since the success of hybrid seed production is dependent upon the genetic purity of the parental lines,
in this study, the experimental F1exp maize hybrid and its parental inbreeds were used as a model
system to examine the discriminative power of morphological, biochemical and SSR markers for
seed purity assay. The highest number of off-type plants was estimated by morphological markers.
According to the comparison of prolamins and albumins banding patterns of parental and derived
F1exp seeds, genetic impurities could not be detected. Molecular analysis detected two types of
genetic profile irregularity. Beside its use for verifying varieties of maize, report on umc1545 primer
pair ability to detect non-specific bands (i.e., off-types), in both the maternal component and F1exp,
which is the first report on this issue yet, strongly supports the recommendation of this SSR marker
use for more accurate and time-efficient maize hybrids and parental lines genetic pyrity testing

AB  - In order to provide continuous progress in maize breeding, it is necessary to expand the genetic base of breeding working collections by introgression of new germplasm into existing heterotic groups. Landraces can be valuable donors of desirable traits despite a large performance gap compared to current commercial varieties. Information on the heterotic pattern of landraces necessary for decision on their incorporation into pre-breeding and breeding programmes, have been made. Molecular characterisation of landraces can provide important information on their diversity before evaluation of their combining ability through crosses to divergent testers. In this study, molecular characterisation of 31 landraces and five divergent testers was performed using 29 polymorphic Simple Sequence Repeat (SSR) markers, in order to examine their genetic divergence. According to cluster analysis, landraces were grouped into four clearly defined groups and one separate branch. The MB1509 landrace differed significantly from all other landraces and testers used. The testers were grouped according to known information on their pedigree. The correspondence analysis pointed out the complexity of mutual relations of landraces and testers. Based on the results obtained by molecular markers, it can be, with high reliability, concluded with which testers the landraces will not give heterosis, making field research more effective (by reducing the number of crosses). The results indicated that the variability of landraces is significant, and that belonging to a certain heterotic group enables the correct selection of the initial material for pre-breeding and their use in breeding programmes.

Grain Yield Predictions of ZP Maize Hybrids

AB  - Maize hybrids are created by crossing inbreds of
opposite heterotic groups, where the discovery of
genetic distances (GD) is of great significance. With
the presence of modern breeding tools for discovering
heterotic pairs and GDs in used material an efficiency in
breeding can be increased for more precisely planned
field work.
For this study 11 crosses were made in order to
compare the prediction of data obtained by GDs
between crossed inbreds with the results of same
hybrids from field testing. Crosses were done in 2020
according to the GD suggestions which ranged from
0.386 to 0.465 to check out is it possible to predict
specific superior hybrid performance before the field
results itself. 25k SNP Ilumina Infinium Array for maize
was used to determine GD values.
As a check ZP 457-market accepted hybrid (registered
in 2018 in Republic of Serbia) was used together with
10 other hybrids. All 11 hybrids shared the same female
component (Lancaster Sure Crop heterotic group),
while male components from opposite heterotic groups
(BSSS and Iowa dent) were used. GD values of used
males ranged from 0.190 to 0.412 clearly stating one
broad heterotic group. The most famous heterotic pair
B-73 x Mo-17 as well as two Institute’s top selling
hybrids (ZP 434 and ZP 606) were also included for
GD comparison with test hybrids with the GD values of
0.488, 0.466 and 0.462 respectively.
Review of B-73 x Mo-17 through ZP 434 and ZP 606
to the newly created hybrids suggests GD values are
decreasing with every new cycle of selection between
crossed material. Results from field testing compared
to GD values show somewhat positive correlation as
for ZP 3 exp which ranked 2nd highest GD value and
3rd rank in grain yield outyielding ZP 457 significantly,
but ZP 457 GD value of 0.458 (ranking third in GD)
suggests higher yielder should be though to find. Grain
yield results proved same hybrid ZP 457 to be one
of the lowest yielders being superior only to ZP 4123
despite high GD value. In top three yielders GD values
were in great correspondence for only one mentioned
combination ZP 3 exp, but for the ZP 4 exp and ZP
457 taking first and third position according to the GD
rank proved to be good only for 5th and 10th rank
within field results, while the lowest yielder ZP 4123
took 4th position according to GD values, having very
high value of 0.457.
This result proves SNP to be powerful tool in predicting
herotic pairs in maize and must be considered for
more precise planned field work. Even though GD
values being so informative on one side they do not

abstract = "Maize hybrids are created by crossing inbreds of
opposite heterotic groups, where the discovery of
genetic distances (GD) is of great significance. With
the presence of modern breeding tools for discovering
heterotic pairs and GDs in used material an efficiency in
breeding can be increased for more precisely planned
field work.
For this study 11 crosses were made in order to
compare the prediction of data obtained by GDs
between crossed inbreds with the results of same
hybrids from field testing. Crosses were done in 2020
according to the GD suggestions which ranged from
0.386 to 0.465 to check out is it possible to predict
specific superior hybrid performance before the field
results itself. 25k SNP Ilumina Infinium Array for maize
was used to determine GD values.
As a check ZP 457-market accepted hybrid (registered
in 2018 in Republic of Serbia) was used together with
10 other hybrids. All 11 hybrids shared the same female
component (Lancaster Sure Crop heterotic group),
while male components from opposite heterotic groups
(BSSS and Iowa dent) were used. GD values of used
males ranged from 0.190 to 0.412 clearly stating one
broad heterotic group. The most famous heterotic pair
B-73 x Mo-17 as well as two Institute’s top selling
hybrids (ZP 434 and ZP 606) were also included for
GD comparison with test hybrids with the GD values of
0.488, 0.466 and 0.462 respectively.
Review of B-73 x Mo-17 through ZP 434 and ZP 606
to the newly created hybrids suggests GD values are
decreasing with every new cycle of selection between
crossed material. Results from field testing compared
to GD values show somewhat positive correlation as
for ZP 3 exp which ranked 2nd highest GD value and
3rd rank in grain yield outyielding ZP 457 significantly,
but ZP 457 GD value of 0.458 (ranking third in GD)
suggests higher yielder should be though to find. Grain
yield results proved same hybrid ZP 457 to be one
of the lowest yielders being superior only to ZP 4123
despite high GD value. In top three yielders GD values
were in great correspondence for only one mentioned
combination ZP 3 exp, but for the ZP 4 exp and ZP
457 taking first and third position according to the GD
rank proved to be good only for 5th and 10th rank
within field results, while the lowest yielder ZP 4123
took 4th position according to GD values, having very
high value of 0.457.
This result proves SNP to be powerful tool in predicting
herotic pairs in maize and must be considered for
more precise planned field work. Even though GD
values being so informative on one side they do not
specifically imply to the solution for detecting the most
superior hybrid combination therefore leaving field
observations to confirm start assumptions.",

AB  - Primary metabolites are directly implicated in plant growth processes, photosynthesis, and
respiration, allowing plants to acclimatize and recover from drought stress. Accordingly, this
study aimed to determine alterations in content of sugars, total protein, seventeen amino acids
and thirty two fatty acids in seeds of maize genotypes differing in drought tolerance in terms
of improved plant productivity e.g. increased grain yield obtained under severe water deficit.
The experiment was conducted under irrigation and rain-fed conditions. Long-term water
deficit stress directly affected plant metabolism, resulting in profound changes in biosynthesis
and transport of evaluated primary metabolites. Water deficit caused the inhibition of sugar
metabolism (i.e. declined level of sucrose, glucose, fructose, maltose and lactose), bein more
pronounced in seeds of drought-sensitive genotypes. Slight decrease in palmitic, linoleic and
α-linoleic fatty acids were evidenced in drougth susceptible and medium drought-tolerant
maize genotypes. However, significant increase was observed for oleic and especially stearic
fatty acid content. In response to water deficit, medium drought-tolerant and tolerant genotypes
exhibited amino acids content increase, especialy for methionine, lysine, isoleucine and
leucine, with preserved incorporation of amino acids into protein chain. This was confirmed
with highly significant positive correlations between total protein and amino acids content
(p≤0.01). The increased protein content in drought tolerant genotypes is linked to altered C-
partitioning, which changes the C/N ratio, to favor more N-assimilation, which was confirmed
with highly significant negative correlations between sugars and protein content (p≤0.01), i.e.
sugars and amino acids content (p≤0.01), respectively. Identification of sensitive sites (i.e.
biochemical pathways for seed reserves) related to seed-filling processes in stressed plants
under long-term water deficit, would provide useful cues in developing strategies to improve
grain yield and its quality.

Multivariate analysis of agronomic traits in mid-season soybean varieties

AB  - Principal Component Analysis (PCA) is a useful tool for processing multiple data, which are
often encountered in breeding practice. This method is suitable for the evaluation of genotypes
on the basis of multiple traits and graphical presentation of relationships between traits. This
study included 16 soybean mid-season genotypes (maturity group I), originated from different
regions of the world, maintained in soybean collection of Maize Research Institute Zemun Polje.
Field trials were carried out at two locations, during two years, according to a RCB design with
three replications. The genotypes were evaluated in respect to eight major agronomic traits: PH –
plant height, NN – node number, PN – pod number, SN – seed number, TSW – 1000 seed weight,
SYP – seed yield per plant, PROT – protein content, OIL – oil content. First two PCA axes
encompassed a large portion of the variance of standardized data (75,9%). Biplot distinguished
genotypes of potential importance for various breeding targets. Two genotypes stood out with the
largest 1000 seed weight. One variety formed a larger number of pods and the seed number per
plant as compared to the group average, achieving the highest grain yield per plant. Two
genotypes were among the most productive ones, with a larger number of pods as well as a
higher 1000 seed weight, compared to the average. The most promising variety was Laura,
which had a high yield and higher protein content than the average, and could be used as a
potential germplasm source for the simultaneous improvement of both traits. Correlations among
traits determined by PC biplot were in accordance with Pearson’s correlation coefficients.

Nutritional and cost effects of adapted quality protein maize on broiler feeding

AB  - ZPQPM13 is a maize hybrid with improved protein quality, adapted to temperate climate growth conditions, developed by crossing a QPM version of ZPL5 commercial line (developed by marker assisted selection) and GS-6 (a QPM line with 50% tropical and 50% temperate germplasm). This hybrid has grain yield as commercial hybrids, over 90% of hard endosperm, contents of tryptophan and protein over 0.075% and 10%, respectively, and stability of tryptophan content over different locations and years. ZPQPM13 was tested for use in broiler feeds, with the aim to reduce utilization of costly synthetic lysine or soybean components. The feeding experiment on Ross 308 broilers was divided into two groups – control (fed with standard maize) and treatment (fed with QPM). In treatment group diets, ZPQPM13 was increased and soybean component decreased for 3%. Chemical analyses revealed higher contents of limiting essential amino acids in mercantile ZPQPM13 compared to standard maize – 0.44 vs. 0.25 % lysine, 0.077 vs. 0.070 % tryptophan and 0.39 vs. 0.27 % threonine. Duration of each feeding trial was 42 days, comprising three phases - starter (1-14 days), grower (15-35 days) and finisher (36-42 days). Feed intake (FI), body mass gain (BMG), average daily gain (ADG) and feed conversion ratio (FCR) were calculated at the end of each phase. In treatment group, FI was reduced, while BMG and ADG were higher in starter phase but lower in grower and finisher phases. FCR was lower in all three phases in treatment group – 1.13 vs. 1.30, 1.25 vs. 1.33 and 1.28 vs. 1.45. Although a small drawback is that final weight was 3.4 % higher in control broilers, comparison of feed and meat prices show that financial benefits of using QPM are still significant compared to using standard maize in feeds, as soybean is four times more expensive than maize.

AB  - Ostrinia nubilalis Hbn. je štetočina kukuruza koja u pojedinim godišnjim dobima može naneti
veliku štetu biljkama. Cilj ovog rada je proučavanje otpornosti odnosno podložnosti hibrida
napadu kukuruznog plamenca i efekata različitih doza insekticida i vremena primene u zaštiti od
ove štetočine na biljkama kukuruza, ali prvenstveno uticaj hibrida na intenzitet napada. Šest
hibrida kukuruza različitih grupa zrenja (ZP 427, ZP 434, ZP 555, ZP 600, ZP 606 i ZP 666)
korišćeno je za proučavanje dejstva napada Ostrinia nubilalis Hbn., na kontrolnu varijantu T1
bez primene insekticida i na varijantu tretiranu sa dva insekticida Fobos EC (bifentrin - 200 ml
ha-1) i kombinacijom Match 050 EC i Nurelle D (lufenuron 50 g l-1 i hlorpirifos 500 g l-1 +
cipermetrin 50 g l-1), 0.75 l ha-1) u dva termina (u prvoj generaciji za vreme leta insekata T2 i
T3 i u drugoj generaciji za vreme leta insekata T4 i T5) u tri ponavljanja. Rezultati su pokazali

AB  - During last years, the total losses from European corn borer (Ostrinia nubilalis Hbn.) can reach
a high threshold of harmfulness although the different agro-technical measures that are being
implemented. According to the sensitivity of hybrids, the damage may vary from only a few
percent in tolerant hybrids, up to 30-40% on moderately intolerant hybrids. The aim of this
study was to evaluate the variability and bioresponsiveness of four maize genotypes (ZP 427,
ZP 434, ZP 600 and ZP 666) to applied insecticides and bioinsecticides and the
interdependence of population dynamics. The research has been conducted in 2016 and 2018
in accordance with the EPPO method in field conditions in maize crops. Out of a total of 11.023
attacks in both years of research, in 2016. the largest number of plants (4.841) were with a
broken stalk above the ear and at the height of the ear and belonged to categories C3 to C8. In
2018. 3.672 plants were dominated by leaf damage and perforation in the stalk type H2.
Analyzing the effectiveness of the tested insecticides and bioinsecticides by application times,
an uniformity in the number of plants with recorded symptoms of damage was observed. Thus,
foliar treatment with chlorantraniliprole insecticide proved to be the most effective with the
least number of attacked plants with a high degree of damage. In contrast, bioinsecticide
treatment of plants, in addition to a large number of infested plants, was dominated by high
categories of damage that deviated from all other treatments as well as from the control. Within
the definition of the degree of tolerance of maize hybrids, the influence of treatment on changes
in secondary metabolites (phytins) in seeds was observed. It was noticed that there is a trend
of positive correlation of the amount of phytin in the seed (mg/g) with the attack of moths of
the first generation and the number of dissected larvae, as well as a trend of negative correlation
with the percentage of attacks of moths of the second generation. Correlation coefficients are
expressed: in the genotype ZP-600, while in the genotype ZP-666 the correlation is the lowest,
ie. irrelevant. The obtained results may be of great importance in future work aimed at the
selection of tolerant genotypes to attack corn flame, taking into account the role of phytin, as
an antioxidant and phosphorus depot in maize grain.

AB  - Plants reconfigure their metabolic pathways to cope with water deficit. The aim of this study was to determine the status of the physiological parameters and the content of phenolic acids in the upper most ear leaf of maize inbred lines contrasting in drought tolerance in terms of improved plant productivity e.g., increased grain yield. The experiment was conducted under irrigation and rain-fed conditions. In drought-tolerant lines, the effect of water deficit was reflected through a chlorophyll and nitrogen balance index increase followed by a flavonols index decrease. The opposite trend was noticed in drought susceptible inbreds, with the exception of the anthocyanins index. Moreover, in comparison to irrigation treatment, opposite trends in the correlations between grain yield and physiological parameters found under water deficit conditions indicated the activation of different metabolic pathways in defense against water deficit stress. Concerning phenolic acid content, water deficit caused the reduction of protocatechuic, caffeic, and sinapic acid in all inbreds evaluated. However, the highly pronounced increase of ferulic and especially cinnamic acid content under water deficit conditions indicated possible crucial role of these secondary metabolites in preventing the harmful effects of water deficit stress, which, in turn, might be useful in maize breeding selection for drought tolerance

Biochemical characterization of soybean genotypes for grain quality improvement

AB  - Soybean is one of the most economically important grain crops. As the
source of plant protein and vegetable oil, it is widely used as food and
industrial crop. Quality of soybean oil is determined by fatty acid (FA)
composition. Biochemical variation of different varieties significantly
facilitates genetic screening of kernel traits and grain quality attributes, thus
leading to one of the most important breeding strategies - improvement of
the grain composition. The main objective of this work was to evaluate
whole kernel biochemical composition of eight soybean varieties and eight
lines from different breeding programs. Phytochemical characterization was
conducted for total protein and oil content, as well as for fatty acids content
(e.g. palmitic, stearic, oleic, linoleic and linolenic acid). According to
Kjedahl method, total protein content ranged from 37.01% to 39.91%. The
obtained results showed that the lowest and the highest values for oil content
were 19.00% and 21.34%, respectively. Gas Chromatography/Flame
Ionization Detector (GC-FID) was used for FA quantification and
determination. The most abundant FA was linoleic acid with a range from
47.45 to 55.7 g/100g, followed by oleic acid with detected rage from 22.59
to 29.07 g/100g of total FA content. Palmitic, linolenic and stearic FA
showed a significantly lower level varied from 9.57 to 11.51 g/100g, 5.42 to
7.18 g/100g and 3.73 to 5.86 g/100g, respectively. This variability observed
in kernel quality among soybean genotypes can provide valuable information
for future breeding.

facilitates genetic screening of kernel traits and grain quality attributes, thus
leading to one of the most important breeding strategies - improvement of
the grain composition. The main objective of this work was to evaluate
whole kernel biochemical composition of eight soybean varieties and eight
lines from different breeding programs. Phytochemical characterization was
conducted for total protein and oil content, as well as for fatty acids content
(e.g. palmitic, stearic, oleic, linoleic and linolenic acid). According to
Kjedahl method, total protein content ranged from 37.01% to 39.91%. The
obtained results showed that the lowest and the highest values for oil content
were 19.00% and 21.34%, respectively. Gas Chromatography/Flame
Ionization Detector (GC-FID) was used for FA quantification and
determination. The most abundant FA was linoleic acid with a range from
47.45 to 55.7 g/100g, followed by oleic acid with detected rage from 22.59
to 29.07 g/100g of total FA content. Palmitic, linolenic and stearic FA
showed a significantly lower level varied from 9.57 to 11.51 g/100g, 5.42 to
7.18 g/100g and 3.73 to 5.86 g/100g, respectively. This variability observed
in kernel quality among soybean genotypes can provide valuable information
for future breeding.",

AB  - Maize Research Institute „Zemun Polje“ (MRIZP) gene bank preserves ex-situ an
active collection (5806 accessions), thus ranking among the five largest European
national maize gene bank collections. Local (i.e. western Balkan) maize landraces
(2217 accessions) are considered to be the most significant genotypes, since they
represent the original biological material created by the process of natural selection
and adapted to local growth conditions, thus offering the great opportunities for
different breeding purposes and contributing to yield increase. Currently, they are
not directly used in breeding, but rather for development of synthetic populations or
core collections for the traits of interest, allowing the integration of the most valuable
traits (adaptability, variability, divergence and heterotic potential). In the last 15
years, in MRIZP gene bank was conducted identification of superior genotypes
regarding specific traits (cytoplasmatic male sterility) and abiotic stress tolerance
(herbicides tolerance). In response to more frequent unpredictable and extreme
climatic events, as well as to growing demand for healthy food and feed, in MRIZP
gene bank were established drought tolerant and improved quality core collections
through implementation of long-term pre-breeding programmes, as the most
promising way of linking genetic resources and commercial breeding programmes.
Moreover, exchange of maize inbred lines from MRIZP gene bank and Agricultural
Institute Osijek working collections, genotyping and phenotyping of inbreds, and
statistical analysis of molecular and agronomic data, is conducted through ongoing
Project “Harmonization of methods for phenotyping, genotyping and management
of genetic resources in maize”. Joint analysis of genetic diversity originated from the
Western Balkan Corn Belt, along with comparative analysis with the same standards
(available genome sequencing data of public lines), will enable the determination of
heterotic pattern, removal of possible duplicate and redundant accessions from the
collections, contributing to a more efficient management of large inbred lines
collections.

AB  - Soybean (Glycine max (L.) Merr.) is one of the most economically important legumes.
As the source of plant protein and vegetable oil it is used as food and industrial crop in
many regions of the world. The genetic base of soybean cultivars is highly narrow,
corresponding to the fact that it is largely a self-pollinated species. Twelve soybean
varieties were evaluated with SSR (Simple Sequence Repeat) markers selected based on
their distribution on the 20 genetic linkage groups. Out of 36 SSR markers, 33 markers
were found polymorphic among analyzed genotypes. Total number of alleles was 88,
ranging between two and four with an average of 2.67 alleles per marker. The
polymorphic information content (PIC) ranged from 0.153 (Satt229, Satt239 and
Satt327) to 0.775 (Satt276). Simple matching similarity coefficient was calculated using
NTSYSpc2 program package. The average genetic similarity coefficient for all pairwise
was 0.57, with highest value (0.84) between Galina and Lela, while the lowest value
(0.46) was found between Bosa and Nena. Dendrogram by the UPGMA (Unweighted
Pair Group Method with Arithmetic Mean) method was constructed on the basis of
genetic similarity matrix. Genotypes were distributed in two groups and one branch,
mostly in accordance with their pedigree.

AB  - Morphological traits are the earliest used markers in germplasm characterization, but their application may be difficult due to the presence of recessive homozygous alleles and their low frequency. Simple Sequence Repeat (SSR) markers are widely used for estimation of genetic diversity within different species, due to their reproducibility, informativeness co-dominant and multi-allelic nature. They are also most effective for evaluation and selection of plant material, as well as assessment of genetic variability and relatedness of maize inbred lines. The aim of our work was to evaluate genetic diversity of maize inbred lines by SSR markers and compare results with their pedigre information. Seventeen polymorphic SSR markers were used to characterized 23 maize inbred lines that belong to diferent breeding programs. A total number of detected alleles was 78 and varied between two to nine, with an average of 4.6 alleles per marker. Based on presence or absence of alleles in each sample coefficient of similarity was calculated by Jaccard in NTSYSpc2 program package, version 2.1. The highest value (0.88) of genetic similarity was calculated between L21 and L22, while the lowest value (0.18) was found between inbred lines L3/L16 and L15/L20. Genetic similarity matrix was used to construct dendrogram by UPGMA (Unweighted Pair Group Method with Arithmetic Mean) method. Dendrogram analysis grouped maize inbred lines in one large cluster with 21 analyzed genotypes and one smaller cluster with two lines. Genetic heterogeneity betweean inbred lines detected by selected set of SSR markers, makes them a good choice for genetic diversity analysis and planning maize breeding programs.

Morphological and physiological response of maize seedlings to chilling stress

AB  - Although accompanied with adverse low temperatures, early maize sowing could be
used to avoid drought during flowering and diminish yield losses. Herein, a small-scale
experiment of low temperature stress (LTS) on maize lines L1 (tolerant), L2 (medium
tolerant) and L3 (susceptible) is presented. Plants were grown in pots exposed to
exterior suboptimal (March) and optimal (late April) temperatures until three leaf stage.
Chlorophyll (CH), flavonoids (FL), anthocyanins (AN) and nitrogen balance (NBI)
indices were measured using Dualex Scientific optical device. Growth parameters were
also determined. Under LTS, number of plants was unchanged for L1 and halved for L2
and L3. Compared to L2 and L3, L1 had significantly higher (p<0.05) shoot fresh
weight (0.649 g vs. 0.406 g and 0.303 g), AN (0.17 vs. 0.13) and FL (1.47 vs. 1.38 and
1.36). For recovery evaluation, plants were transplanted into the field. Transplanted
stressed L1 plants showed the highest grain yield per plant (55g) in the field. Due to
high correlations (p<0.01) between FL in three leaf stage and grain yield per plant, FL
could be used as an indicator of plant recovery of maize genotypes exposed to LTS
during early sowing.
AB  - Ranom setvom kukuruza se može izbeći efekat suše u fazi cvetanja i tako preduprediti smanjenje
prinosa uprkos nepovoljnim temperaturama u tom periodu. U ovom istraživanju prezentovani su
rezultati efekta niskih temperatura na tri linije kukuruza: L1 (tolerantna), L2 (srednje osetljiva) i
L3 (osetljiva). Biljke su gajene do faze trećeg lista u saksijama izloženim suboptimalnim (mart) i
optimalnim (april) spoljnim temperaturama. Sadržaj hlorofila, flavonoida i antocijana kao i
nitrogen balance index (NBI) su mereni korišćenjem uređaja Dualex Scientific (Force-A, Orsay,
France). Takođe, mereni su i parametri rasta. U poređenju sa L2 i L3, L1 je imao značajno veću
(p<0.05) svežu masu nadzemnog dela biljke (0.649 g vs. 0.406 g i 0.303 g), antocijana (0.17 vs.
0.13) i flavonoida (1.47 vs. 1.38 i 1.36). Biljke su presađene u polje radi procene oporavka.
Presađene biljke genotipa L1 su pokazale najveći prinos po biljci u polju (55g). S obzirom na
visoku korelaciju između sadržaja flavonoida u fazi trećeg lista i prinosa po biljci (p<0.01),
flavonoidi mogu biti korišćeni kao indikator oporavka biljke kukuruza izloženih niskim
temperaturama u fazi rane setve

nitrogen balance index (NBI) su mereni korišćenjem uređaja Dualex Scientific (Force-A, Orsay,
France). Takođe, mereni su i parametri rasta. U poređenju sa L2 i L3, L1 je imao značajno veću
(p<0.05) svežu masu nadzemnog dela biljke (0.649 g vs. 0.406 g i 0.303 g), antocijana (0.17 vs.
0.13) i flavonoida (1.47 vs. 1.38 i 1.36). Biljke su presađene u polje radi procene oporavka.
Presađene biljke genotipa L1 su pokazale najveći prinos po biljci u polju (55g). S obzirom na
visoku korelaciju između sadržaja flavonoida u fazi trećeg lista i prinosa po biljci (p<0.01),
flavonoidi mogu biti korišćeni kao indikator oporavka biljke kukuruza izloženih niskim
temperaturama u fazi rane setve",

Marker assisted selection for β-carotene rich maize: two-level selection procedure in BC2 generation

AB  - This paper is a part of the breeding program conducted at the Maize Research Institute "Zemun Polje", with the aim to convert the standard maize to β-carotene rich genotypes adapted to temperate regions using the marker assisted selection. When SSR marker specific to the crtRB1 gene has been tested on parental lines, distinct polymorphism was observed between the donor and recurrent parents. The same marker has also been validated in BC1 generation when heterozygous individuals were clearly distinguishable from the homozygous dominants. The results presented herein refer to two-level selection procedure in BC2 generation. First, BC2 plants were analyzed with crtRB1-specific molecular marker to separate heterozygotes from dominant homozygotes. Percentage of heterozygous plants was approximately 50%, which was in accordance with the expected Mendelian ratio of 1:1. Second, the selected heterozygotes were screened with 30 polymorphic SSR markers distributed throughout the maize genome to identify genotypes with the highest recovery of recurrent parent’s genome (RPG). The RPG values among three analyzed parental lines and their respective progenies ranged from 85-99%. For each line separately, RPG values were: 86-97% (RP1), 90-95% (RP2) and 85-99% (RP3). Plants with RPG above 95% were selfed to produce BC2F2 generation in which homozygous recessive individuals would be identified. Those genotypes will be subjected to biochemical and phenotypic evaluation to confirm their nutritional and agronomical superiority. Finally, these β-carotene enriched lines, as well as the resulting improved hybrids, can be used in the biofortification programs.

AB  - Initial studies related to cytoplasmic male sterility (CMS) were performed by Rhoeds in 1931. 
CMS is used in maize to enhance efficiency of seed production with simultaneous cost 
redaction. The majority of studies showed positive effects of CMS on maize grain yield. 
Grain yields recorded male sterile plants were higher by 5-10% than the ones in female fertile 
plants. Seven maize inbred lines of different origin and growing season were analysed. Each 
inbred was analysed in five variants: original inbred (N), CMS-C, RfC, CMS-S and RfS. The 
aim of this study was to compare grain yields and yield components of original inbreds and 
their CMS and Rf variants. The highest yields of observed inbreds (5.303 and 5.197 t/ha) 
were recorded in those with C and S cytoplasm, respectively. According to the LSD test, at 
significance levels of 0.05 and 0.01, the longest ears of 15.57, 15.56 and 15.46 cm were 
detected in inbreds with original, S and C cytoplasm, respectively. The highest kernel row 
number (12.98) at both significance levels was established in inbreds with C cytoplasm. The 
highest number of kernels per row (33.92) at both levels of significance was recorded in 
inbreds with S cytoplasm. The greatest kernel depth (0.8212 and 0.8196 cm) at both 
significance levels was established in inbreds with C and S cytoplasm, respectively. The 
greatest 1000-kernel weight at both levels of significance was detected in inbreds with normal 
cytoplasm. The highest number of kernels per m2
(2716 and 2676) was recorded in inbreds 
with C and S cytoplasm, respectively.

AB  - Soybean (Glycine max (L.) Merr.) is one of the oldest crop in the world, with a history of growing over 4000 years. At the same time it is the most important legume in the global world production. Simple Sequence Repeat (SSR) markers are widely used for estimation of genetic diversity within different species, due to their reproducibility, co-dominant and multi-allelic nature. Molecular characterization among soybean genotypes was done with 36 SSR markers, with at least one primer from each linkage group, to determine the genetic diversity. Twenty soybean accessions and varieties obtained from Maize Reasearch Institute „Zemun Polje“ were analyzed. A total number of detected alleles was 116 and number of alleles varied between two to six, with an average of 3.22 alleles per marker. Thirteen unique alleles were obtained from eleven SSR loci in nine genotypes. Molecular analysis revealed a high polymorphism of SSR loci. Pairwise coefficients of genetic similarity calculated by Dice ranged from 0.14 to 0.74. Genetic similarity matrix was used to construct dendrogram by UPGMA (Unweighted Pair Group Method with Arithmetic Mean) method in NTSYSpc2 program package. This clustering method distinguished 20 soybean genotypes in two subclusters and one branch.
